摘要
【目的】观察过敏性及非过敏性哮喘发病过程中2型固有免疫细胞(ILC2)的差异,探讨ILC2与哮喘临床评价指标的相关性。【方法】研究对象来自我院2017年6月至2018年1月的过敏性和非过敏性哮喘患者,收集相应临床基线资料并评估病情、进行血常规、肺功能等检查;应用流式细胞术检测外周血ILC2;通过ELISA和多因子检测分别测定总Ig E及s Ig E和Th2型细胞因子。【结果】哮喘(Asthma)组的ILC2比例和计数与嗜酸性粒细胞(EOS)计数及s Ig E正相关(r=0.37,0.32,0.56,0.57;P<0.01~=0.04);过敏性哮喘(AA)组的ILC2比例和计数为0.04(0.03~0.11)%和338(255~778)cells/m L,均高于非过敏性哮喘(NAA)组的0.01(0.01~0.02)%及89(0~169)cells/m L(P<0.01);中/重度患者ILC2比例为(0.10±0.08)%,高于间歇/轻度患者的(0.04±0.01)%(P=0.04);AA组中ILC2比例和计数与FEV1/FVC呈负相关(r=-0.61,-0.56;P<或=0.01)。【结论】ILC2参与过敏性哮喘的发病机制,与EOS表型、过敏炎症反应和病情严重程度相关,可作为过敏性哮喘的有效评价指标之一,为过敏性哮喘的治疗提供新方向。
【Objective】To observe the difference of type 2 innate lymphoid cells(ILC2)in allergic and non-allergicasthma,and to investigate the relationship between ILC2 and other major clinical parameters.【Methods】Asthma patientscame from the First Affiliated Hospital of Sun Yat-sen University from June 2017 to January 2018. Clinical baseline datawere collected and levels of asthma severity were assessed. Besides,blood routine and lung function were examined. ILC2 were detected via flow cytometry. Total Ig E,s Ig E,and Th2 cytokines were quantitated via ELISA and Multiple Cyto-kine Detection System,respectively.【Results】The percentage of ILC2 in lymphoid cells and the number of ILC2 were cor-relate with EOS and s Ig E positively,respectively(r=0.37,0.32,0.56,0.57;P〈0.01~=0.04). The percentage and thenumber of ILC2 in allergic asthma group were 0.04(0.03~0.11)% and 338(255~778)cells/m L,which were both higherthan 0.01(0.01~0.02)% and 89(0~169)cells/m L in non-allergic asthma group(P〈0.01). The percentage of ILC2 was(0.10±0.08)% in moderate and severe patients,which was more than(0.04±0.01)% in intermittent and mild patients(P=0.04). The percentage and the number of ILC2 were correlate with FEV1/FVC negatively(r=-0.61,P〈0.01;r=-0.56,P=0.01).【Conclusions】ILC2 participate in pathogenesis of allergic asthma and correlate with EOS,allergy and level of asth-ma severity,which would be an evaluation indicator of allergic asthma and provide a new direction in asthma therapy.
