隐丹参酮对荷瘤小鼠化疗后免疫功能低下的影响 被引量:3

摘要 目的探讨隐丹参酮(CPT)对荷瘤小鼠顺铂(DDP)化疗后免疫功能低下的影响。方法40X小鼠分为5组,每组8只,Pc9细胞建移植瘤动物模型,分别为空白对照组组、模型组、CPT组(100mg/kg)、DDP组(2mg/kg)及联合用药(100mgCPT+2mg/kgDDP)组,隐丹参酮连续给药15d,顺铂2次/周给药(每周第1天、第4天),停药24h后,测脾脏指数、CCK8检测T细胞增殖率、流式细胞术检测T细胞亚群、ELISA法检测荷瘤小鼠血清中IFN-1、IL-2的含量。结果与空白对照组比较,模型组免疫功能受到明显抑制(P〈O.01);与模型组比较,CPT可明显提高荷瘤小鼠脾脏指数、T细胞增殖率、CD3^+、CD8^+、NK细胞、IFN-1、IL-2的表达(P〈001),而DDP的作用相反(P〈O01或P〈O.05);与顺铂组比较,CPT组、联合用药组脾脏指数、T细胞增殖率、CD3^+、NK细胞、IFN-γ、IL-2表达明显上升(P〈001),而CD4^+、CD8^+、CD4^+/CD8^+变化不明显(P〉005);与隐丹参酮组比较,联合用药组脾脏指数、T细胞亚群、IL-2等差异无统计学意义(P〉0.05),T细胞增殖数量、IFN-γ差异有统计学意义(P〈O.01或P〈0.05)。结论隐丹参酮可提高化疗后机体的免疫功能,其机制可能与隐丹参酮提高血清中IFN-1、IL-2含量相关,对机体的免疫脏器也有-定的保护作用,具有-定的临床应用前景。 Objective To study the immunodeficiency of cryptotanshinone ( CPT ) on post-chemotherapeutic tumor-bearing mice. Methods Lung adenocarcinoma PC9 cell transplanted tumor animal models were divided into five groups: control group, model group, CPT group ( 100mg/kg ) , DDP group (2mg/kg) , and the combination group of CPT and DDP ( 100mg CPT+2mg/kg DDP ) , each group had eight mice. Cryptotanshinone was administered continuously to 15d, cisplatin 2 times / week (first days, fourth days per week ) . After the withdrawal of 24h, the spleen index, the proliferation rate of T cells were measured by CCKS, the T cell subgroup was detected by flow cytometry, and the content of IFN- gamma and IL-2 in the serum of the tumor bearing mice was detected by ELISA. Results Compared with the blank control group, the immune function of the model group was significantly inhibited (P〈0.01) . Compared with the model group, the spleen index, T cell proliferation rate, CD3+, CD8+, NK cells, IFN- gamma and IL-2 expression were significantly increased by CPT in the tumor bearing mice ( P〈0.01 ) , but the effect of DDP was opposite (P〈0.01 or P〈0.05 ) . Compared with cisplatin group, the expression of spleen index, T cell proliferation rate, CD3+, NK cell, IFN- gamma and IL-2 in CPT group and combined drug group increased significantly ( P〈0.01 ) , while CD4+, CD8+ and CD4+/CD8+ were not significant (P〉0.05) . Compared with the cryptotanshinone group, there was no significant difference in the spleen index, T cell subgroup and IL-2 (P〉0.05) in the combination group (P〉0.05) . The number of T cells and the difference of IFN- gamma were statistically significant (P〈0.01 or P〈0.05 ) . Conclusion CPT improves immune function after chemotherapy, and its mechanism may be related to the increase of the content of IFN- gamma and IL-2 in serum by cryptotanshinone. It also has a certain protective effect on the immune organs of the body, and has a certain clinical application prospect.
