中国水仙NtPLATZ1抗血清的制备及效价分析

Antiserum Preparation and Titer Analysis of NtPLATZ1 in Narcissus tazetta var.chinensis

将本试验前期成功表达的中国水仙锌指蛋白Nt PLATZ1基因融合蛋白免疫大白兔,收集抗血清,制备多克隆抗体。用酶联免疫吸附测定法(ELISA)检测其效价,通过免疫印迹检测(western-blot)分析该血清的特异性。ELISA检测得出多克隆抗体的效价为l∶6 400,Western blotting结果表明,制备的抗血清能与诱导表达的融合蛋白发生特异性反应。免疫大白兔获得的血清特异性强,效价高,可用于Nt PLATZ1蛋白功能的研究。

Rabbits were immunized with the recombinant protein of NtPLATZ1 expressed in Escherichia coli to prepare polyclonal antibody, The antiserum was collected and evaluated by ELISA method and the specificity of antiserum was detected by western-blot. The result showed that the antiserum had a good immunoactivity at a dilution of 1：6 400. The western-blot analysis showed that the serum obtained from immunized rabbits has strong specificity and high titer, and can be used to study the function of NtPLATZlgene from Narcissus tazetta var. chinensis.
