摘要
目的:利用Illumina Miseq测序技术探究病理黄腻苔、生理黄腻苔及健康薄白苔之间舌苔菌群门与属水平上的差异性。方法:采集病理黄腻苔、生理黄腻苔和健康薄白苔样本各50例,归为3组。对3组舌苔样本提取总DNA,并筛选出12份病理黄腻苔样本、12份生理黄腻苔样本及11份健康薄白苔样本,共35份,以Illumina Miseq PE300为测序平台进行高通量测序,并对测序数据进行处理和分析。结果:对35份样本的测序数据进行统计后共得到1 282 089条优质序列,全部样本的平均序列读长为443.35bp。与数据库进行比对后确定分属于12个门、23个纲、39个目、66个科、133个属、246个种的细菌。3组样本的群落构成有一定相似性,在门水平上有厚壁菌、变形菌、梭杆菌、放线菌和拟杆菌5种相同优势菌门;在属水平上有普雷沃菌-7、奈瑟氏菌、韦荣球菌和链球菌等13种优势菌属。但3组样本在门、属水平上优势菌群组成存在差异。结论:变形菌、放线菌、奈瑟氏菌、韦荣氏菌和嗜血杆菌可能是导致黄腻苔形成并区别于健康舌苔的菌群,同时变形菌、奈瑟氏菌、韦荣球菌可能是区分生理与病理黄腻苔的关键菌种,而生理黄腻苔与健康舌苔在群落结构上无显著差异。
Objective: To explore the flora differences in phylum and genus level between pathological yellow-greasy coating, physiological yellow-greasy coating and healthy thin-white coating by using Illurnina Miseq sequencing technology. Methods: Collected pathological yellow-greasy coating, physiological yellow-greasy coating and healthy thin-white coating samples each 50 cases, divided into three groups. Tongue samples extracted total DNA of three groups, and total 35 samples were selected to make high throughput sequencing with Ulumina Miseq PE300, as 12 samples of pathological yellow-greasy coating, 12 samples of physiological yellow-greasy coating and 11 samples of healthy thin-white coating, then processing and analysis the data. Results: After sequencing the data from 35 samples, 1 282 089 high-quality sequences were obtained, and the average reading length of all samples was 443.35bp. Compared with the bacterial database, the bacteria were identified as 12 phylums, 23 classes, 39 orders, 66 families, 133 genera and 246 species. The community composition of the three groups was similar and showed the same dominant species at the phylum and genera level, however, there were differences in the proportion of dominant populations between the 3 groups. Conclusion: Proteobacteria, Actinobacteria, Prevotella, Neisseria, Veillonella and Haemophilus may lead to the formation of yellow-greasy coating and different from healthy coating, Actinobacteria and Prevotella may be distinguished with yellow-greasy coating key strains of healthy thin- white coating, and no significant differences in community structure between physiological yellow-greasy coating and health coating.
