摘要
目的:探究脊髓中肿瘤坏死因子α(TNF-α)在加味升降散治疗神经病理性疼痛中的作用机制。方法:将实验大鼠随机分为5组:分别为假手术组,模型组,加味升降散低、中、高剂量组。模型组和治疗组构建大鼠右侧改良坐骨神经慢性缩窄性损伤(i CCI)经典动物模型,并以术后第7天开始灌胃不同剂量加味升降散,分别在给药第1、7、14、21天处死大鼠,抽取脊髓样本检测TNF-α指标在mRNA和蛋白的表达量。结果:给药后,各组脊髓中TNF-α的mRNA及蛋白表达量总体呈下降趋势。加味升降散高剂量组脊髓中TNF-α的mRNA的表达量在给药后第1天即显著低于模型组,至第7天到第21天,下降趋势不明显,趋于平衡;加味升降散低、中剂量组于给药第7天显著低于模型组,之后趋于平衡。加味升降散高、中剂量组脊髓中TNF-α的蛋白表达水平于给药第1天显著低于模型组,之后仍明显下降。加味升降散低剂量组中TNF-α的蛋白表达量在给药后不同治疗时间点,与模型组无明显差异。结论:加味升降散可下调脊髓中TNF-αmRNA和蛋白的表达量,其可能是治疗和缓解NP的机制之一。另外,高剂量显示较好的治疗效果,低剂量无明显治疗效果。
Objective: To investigate the mechanism of TNF- a in the spinal cord of SD rats with neuropathic pathological pain were treated by Jiawei Shengjiang Powder. Methods: The rats were randomly divided into five groups including the sham operation group, the model group, the Jiawei Shengjiang Powder treatment group, like tow dose group, middle dose group and high dose group. The classic animal model of modified sciatic nerve chronic constrictive injury (iCCI) was established in fight side of rats which belong to the model group and treatment group, the rats were gavaged different dose of Jiawei Shengjiang Powder on the 7th day after surgery, and the rats were sacrificed on the 1st day, the 7th day, the 14th day and the 21st day to take spinal cord samples to detect the expression of TNF- c~ in mRNA and protein. Results: The mRNA and protein expression of TNF- a in the spinal cord were decreased in different group after treatment. The mRNA expression of TNF- a in the spinal cord in the high group was significantly lower than that in the model group on the 1st day after treatment, but the downward trend was not obvious and tend to balance at 7th day to the 21st day. The low dose and medium dose were significantly lower than the model group at the 7th day, then it was tended to the balance. The protein expression level of TNF- a in spinal cord of high dose and middle dose group were significantly lower than that of model group on the 1st day, and then decreased significantly. The protein expression level of TNF- a in the spinal cord of low dose group was not significantly different from that in the model group at different time points after treatment. Conclusion: The mRNA and protein expression of TNF- a in the spinal cord was down-regulated during the treatment by Jiawei Shengjiang Powder, which was one of the mechanisms of treating and alleviating neuropathic pathological pain. In addition, the high dose showed better therapeutic effect and low dose had no significant treatment effect.
