益气养阴祛瘀方对干燥综合征患者血清TLR-IFN-BAFF信号通路的影响

Effects of Yiqi Yangyin Quyu Formula on TLR-IFN-BAFF signaling pathway in patients with primary Sjogren syndrome

目的:通过观察干燥综合征(SS)患者治疗前后血清Toll样受体9(TLR9)、α干扰素(IFN-α)、B细胞刺激因子(BAFF)蛋白的表达改变,探讨益气养阴祛瘀方对SS患者血清TLR-IFN-BAFF信号通路的影响。方法:将60例女性SS患者随机分为中药组(口服益气养阴祛瘀方水煎剂)和西药组(口服硫酸羟氯喹片),每组各30例,连续治疗24周;另选取20名女性健康体检者作为健康对照组。观察中药组和西药组治疗前后的临床症状评分及血沉(ESR)、血清IgG含量的变化;并通过酶联免疫吸附测定法(ELISA法)检测60例患者治疗前后及20名健康人血清TLR9、IFN-α、BAFF蛋白水平。结果:与健康对照组比较,治疗前两组血清TLR9、IFN-α、BAFF蛋白水平均显著升高(P<0.05);与本组治疗前比较,治疗后两组各项临床症状评分、ESR、血清IgG及血清TLR9、IFN-α、BAFF蛋白水平均显著降低(P<0.05)。结论:益气养阴祛瘀方可以改善SS患者的临床症状,其治疗机制可能与影响SS患者体内TLR-IFN-BAFF信号通路的表达有关。

Objective： To study the effects of Yiqi Yangyin Quyu Formula on serum TLR-IFN-BAFF signaling pathway in SS patients through observing the changes of serum Toll-like receptor 9 （TLR9）, interferon-alpha （IFN-α ）, and B-cell stimulating factor （BAFF） proteins before and after treatment in patients with Sjogren syndrome （SS）. Methods： Sixty patients with SS were randomly divided into Chinese medicine group （oral administration of Yiqi Yangyin Quyu Formula water decoction） and western medicine group （oral administration of hydroxychloroquine sulfate tablets）. Thirty patients in each group were treated for 24 consecutive weeks, and 20 healthy subjects were selected as the healthy control group. The scores of clinical symptoms, ESR, and serum IgG levels before and after treatment in the Chinese medicine group and the western medicine group were observed; the serum TLR9, IFN-α and BAFF protein levels were measured in 60 patients before and after treatment and in 20 healthy individuals by immunosorbent assay （ELISA）. Results： Compared with the healthy control group, serum TLR9 protein, IFN-α protein and BAFF protein levels in the Chinese medicine group and the western medicine group before treatment were significantly increased （P〈0.05）; compared with the levels before treatment, clinical symptom scores, ESR, serum IgG, serum TLR9 protein, IFN-a and BAFF protein levels in the Chinese medicine group and the western medicine group after treatment were significantly lower （P〈0.05）. Conclusion： yiqi Yangyin Quyu Formula can improve clinical symptoms of SS patients, the treatment mechanism may be related to expression of TLR-IFN-BAFF signaling pathway in SS patients.
