旋转蒸发器制备川芎挥发油羟丙基-β-环糊精包合物的Box-Benhnken-响应面法优化 被引量:11

Optimization of Rotatory Evaporator Inclusion Process of Volatile Oil from Chuanxiong Rhizoma and Hydroxypropyl-β-cyclodextrin by Box-Benhnken Design-response Surface Methodology

摘要 目的采用Box-Benhnken-响应面法优化川芎挥发油羟丙基-β-环糊精(HP-β-CD)包合物的制备工艺。方法采用溶液法,仅用旋转蒸发器制备川芎油HP-β-CD包合物,以包合物含油率及挥发油包合率为评价指标,采用Box-Benhnken试验考察HP-β-CD与川芎油的投料质量比、包合时间及包合温度对包合工艺的影响,对试验数据进行多元线性回归和二项式方程拟合,通过响应面法优选包合工艺条件并进行验证试验。结果最佳包合工艺条件为HP-β-CD与川芎油的投料质量比8g·g-1,包合温度55℃,包合时间2h;包合物含油率及挥发油包合率预测值与理论值的偏差较小。结论 Box-Benhnken-响应面法适用于优化旋转蒸发器制备川芎油-HP-β-CD包合物的制备工艺,建立的数学模型具有良好的预测性。 Objective To optimize the preparation process of inclusion complex of volatile oil from Chuanxiong Rhizoma and hydroxypropyl-β-cyclodextrin(HP-β-CD). Methods Taking composite score of inclusion rate of volatile oil and inclusion compound oil content,Box-Benhnken design-response surface methodology was adopted to optimize inclusion process with inclusion temperature,reaction time,ratio of HP-β-CD and volatile-oil as factors,with rotary evaporatoronly. Results The best process was as follows,ratio of HP-β-CD and volatile-oil was 8: 1,inclusion temperature was 55℃,inclusion time was 2 h. The oil content of inclusion compound and the inclusion ratio of volatile oil have little deviation from the theoretical value. Conclusion Each index of the optimized hydroxypropyl-β-cyclodextrin complex was well fitted with the predicted value. The adopted model was reasonable and effective.
