摘要
目的:建立具有免疫功能的小鼠原位荷瘤模型,并优化其评价方式。方法:通过注射绿色荧光蛋白(GFP)标记的Lewis肺癌细胞(LLC)悬液至C57BL/6小鼠肺部。方法:利用表达GFP基因的病毒转染Lewis肺癌细胞。采用肿瘤细胞悬液注射法建立Lewis肺癌原位模型。小动物活体成像系统鉴定其成瘤情况。CT薄层扫描和HE染色确定其位置和病理性质。结果:在荷瘤2周之后CT显示肺部有占位,小鼠胸部可观察到绿色荧光信号,HE染色确定其为肿瘤组织。结论:GFP标记的Lewis肺癌小鼠原位模型的建立和评估是可行的,为肺癌的基础研究提供了有效的动物模型。
Objective:To establish an orthotopic mouse model with normal immune function,and to optimize the evaluation system for this model.Methods:Lewis lung carcinoma cells were transfected with the GFP lentiviruses,then,these GFP-labeled cancer cell suspension was orthotopically injected into the lungs of C57 BL/6 mice.The growth of the orthotopic tumor was analyzed by an in vivo fluorescence imaging system for small animal.Thin-Layer Scanning CT was used to identify the tumor loci,and pathological characteristics were examined by hematoxylin and eosin(HE)staining.Results:Occupying lesion was observed on the images of CT scan 2 weeks after orthotopic injection.Green fluorescence was visible through the flap,and cancer was confirmed by HE staining.Conclusion:The establishment and evaluation of GFP-labeled orthotopic Lewis lung cancer mouse model is feasible.It provides an effective platform for basic research of lung cancer.
