bFGF与VEGF在骨折愈合过程表达及促进愈合机制分析 被引量:7

摘要 目的探计碱性成纤维细胞生长因子(bFGF)对骨折愈合过程中血管内皮生长因子(VEGF)表达的影响及二者在促进骨折愈合中的作用。方法将407,SD雄性大鼠随机均分为2组,手术建立胫骨骨折模型,其中观察组骨折处局部注射bFGF,而对照组骨折处局部注射生理盐水,两组分别于术后D7、D14、D21、D28随机处死5R大鼠,采集骨折断端的骨痂及周围组织进行染色,免疫组织化学染色测定VEGF表达新生血管量,ELISA法检测vEGF表达水平。结果术后D7、D14、D21、D284个时间段骨折处新生血管量:D7两组均未见新生血管形成。D14对照组新生血管量为0,观察组为(14.46±2.68),差异有统计学意义(P〈0.05)。D21对照组新生血管量为(1263±2.21),观察组为(17.10±2.53),差异有统计学意义(P〈005)。D28对照组新生血管量为(28.39±4.52),观察组为(42.32±5.82),差异有统计学意义(P〈0.05)。术后D7、D14、D21、D284个时间段观察组VEGF表达量均高于对照组,差异有统计学意义(P〈0.05)。结论骨折处局部注射bFGF可以增加骨折愈合过程中VEGF表达量,并延长其表达时限,两者共同作用可以促进骨折愈合过程。 Objective To investigate the effect of during fracture healing and the role of both in promoting fracture healing. Methods 40 male Sprague-Dawley rats were randomly divided into 2 groups. The tibial fracture model was established by surgery. Bone fibroblast growth factor ( FGF ) was injected locally in the fracture of the experimental group, while saline was injected locally in the fracture of the control group. Five rats were randomly sacrificed at D7, D14, D21 and D28 after operation. The callus of the fracture and the surrounding tissue were harvested for staining. Immunohistochemical staining was used to determine the amount of vascular endothelial growth factor ( VEGF ) expression. The vascular endothelial growth was detected by ELISA factor expression level. Results There were no neovascularization in the D7, D 14, D21 and D284 fractures at the time points after operation. The number of neovascularization in D14 control group was 0 and in experimental group was ( 14.46 ± 2.68 ) , the difference was statistically significant. The number of neovascularization in D21 control group was ( 12.63 ±2.21 ) and in experimental group was ( 17.10 ±2.53 ) , the difference was statistically significant. The number of neovascularization in D28 control group was ( 28.39 ± 4.52 ) and in experimental group was ( 42.32 ± 5.82 ) , the difference was statistically significant. The expression of VEGF in experimental group after D7, D14, D21 and D284 were higher. Conclusion Local injection of basic fibroblast growth factor VEGF prolongs the expression of vascular endothelial growth factor ( VEGF ) in the process of fracture healing. Both of them can promote fracture healing.
