摘要
以片状载体作为固定化载体,戊二醛为交联剂,制备固定化胰蛋白酶。以固定化时间、固定化温度、固定化pH值和固定化酶质量分数对固定化效果进行评估。结果表明,在固定化时间1.5 h,固定化温度40℃,固定化pH值8.0,胰蛋白酶质量分数2%时,固定化效果最好,回收率最高。在最佳固定化条件下,固定化酶活力回收率可达46.89%,固定化胰蛋白酶表现出较好的热稳定性和酸碱稳定性。
Immobilized trypsin was prepared by using flaky carrier as carrier and glutaraldehyde as cross-linking agent. Be- sides, an assessment had been made on fixed time, fixed pH value and immobilized enzyme concentration. The results indi- cated that the immobilization temperature was 40℃, immobilization pH value was 8.0, the trypsin concentration was 2% when the immobilization time was 1.5 h, achieving the best immobilization effect and the highest recovery rate. Under the opti- mum immobilization conditions, the recovery rate of immobilized enzyme reached 46.89%, showing good thermal stability and acid base stability of the immobilized trypsin.
