摘要
目的建立黄芩茎及叶中野黄芩苷的含量测定方法。方法采用高效液相色谱(HPLC)法,C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.02%甲酸(28∶72)为流动相等度洗脱,流速1.0 m L·min-1,检测波长335 nm。结果野黄芩苷在0.138 0~1.657 0μg范围内与峰面积呈良好的线性关系,r=0.999 8,平均回收率为98.24%,RSD为0.96%;黄芩茎的野黄芩苷含量为0.86%,黄芩叶的野黄芩苷含量为2.83%。结论该法简单方便,可用于测定黄芩茎及叶野黄芩苷的含量,对深入研究黄芩茎叶化学成分生物合成机理,确定黄芩茎叶药材最佳采收期以及制定黄芩茎叶药材质量标准提供理论依据。
Objective To establish a method for the determination of scutellarin in Scutellaria baicalensis stems and leaves.Methods The separation by HPLC was performed on C18( 4.6 mm×250 mm,5 μm),the mobile phase was methanol-0.02% formic acid solution( 28 ∶ 72),with flow rate was 1.0 m L·min-1 and the detection wavelength was 335 nm.Results The content of scutellarin within the range between 0.138 0 ~ 1.657 0 μg had a good linear relationship with peak area( r= 0.999 8).The average recovery was 98.24%,with RSD = 0.96%; The content of scutellarin in the stems of Scutellaria baicalensis was 0. 86% and the content of scutellarin in the leaves of Scutellaria baicalensis was 2. 83%. Conclusion The method was simple and convenient,and it can be used to determine the content of scutellarin from medicinal herbs like stems and leaves of Scutellaria baicalensis.It provided theoretical basis for the in-depth study of biosynthesis mechanism of chemical constituents from Scutellaria baicalensis,the best harvest time for Scutellaria baicalensis stem-leaf,and quality standards of Scutellaria baicalensis stem-leaf herbs.
