摘要 目的研究促红细胞生成素衍生肽HBSP对顺铂诱导急性肾损伤的保护作用以及作用机制。方法 65只健康雄性清洁级SD大鼠,随机分为对照组,顺铂组、EPO组、HBSP组。分布检测大鼠血肌酐、尿素、血常规以及肾小管上皮细胞凋亡、肾脏组织TNF-α、NF-κB表达水平、尿Kim-1、NGAL水平。结果 EPO组与HBSP组血肌酐和尿素、尿Kim-1与NGAL以及肾组织Tunel染色阳性百分比、免疫组化染色TNF-α、NF-κB肾脏表达水平较对照组升高,与顺铂组相比均显著下降。各组红细胞计数和血红蛋白与对照组比较差异无统计学意义。结论 HBSP对顺铂诱导的急性肾损伤具有保护作用,其机制可能与抑制炎性反应以及细胞凋亡有关。 Objective To investigate the protective effect and mechanism of helix B surface peptide (HBSP) on cisplatin-induced aucte kidney injury in SD rats. Methods SD rats were randomly divided into 4 groups:control group, cisplatin group, the EPO group and HBSP group. Serum creatinine, urea, red blood cell count, hemoglobin and urine Kim-1, NGAL were detected as well as the expression of TNF-α, NF-κB in renal tissues. Apoptosis of tubular epithelium was observed by TUNEL assay. Results The levels of serum creatinine, urea and urine KIM-1, NGAL were decreased, accompanied with significantly less Tunel-positive cells, as well as less expressins of TNF-α and NF-κB in EPO group and HBSP group. Both red blood cell counts and hemoglobins in all groups were no significant difference. Conclusion HBSP enhance recovery from cisplatin-induced AKI in rats by its inhibition of inflamation and tubular epithelium apoptosis.
