摘要
目的探讨金虎退黄汤对α-萘异硫氰酸酯(ANIT)诱导的大鼠急性肝内胆汁淤积的干预作用及作用机制。方法将21只SD大鼠随机分为空白组、模型组、实验组,每组7只,按规定时间点进行造模、给药。末次造模后48 h,检测大鼠血清中总胆红素(TBil)、直接胆红素(DBil)、总胆汁酸(TBA)、丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)、谷氨酰转肽酶(GGT)、碱性磷酸酶(ALP)含量。取血后,处死大鼠,摘取心、肝、脾、肺、肾,称定质量,计算脏器指数;肝组织做组织切片HE染色,光镜观察肝组织病理变化。结果各组大鼠体质量虽缓慢增加,但变化无明显差异(P>0.05);实验组血液生化指标TBil,DBil,TBA,AST,ALP下降明显(P<0.05),病理组织切片改善较显著,但ALT和GGT下降不明显(P>0.05)。结论金虎退黄汤对胆汁淤积性肝损伤大鼠具有显著的退黄、降酶、修复肝细胞损伤作用,可能通过抑制炎性反应及肝细胞坏死发挥作用。
Abstract: Objective To investigate the intervention effect of Jinhu Tuihuang Decoction on acute intrahepatic cholestasis induced by a - Naphthylisothiocyanate(ANIT) in rats and its mechanism. Methods Totally 21 SD rats were randomly divided into the blank group, model group and experimental group, 7 rats in each group, the rats were modeled and administrated according to the prescribed time points. 48 h after the last modeling, the levels of serum TBil, DBil, TBA, ALT, AST, GGT, ALP were detected. After taking blood, the rats were sacrificed, and the heart, liver, spleen, lung and kidney were taken out and weighed , the visceral indexes were measured , the liver tissue was stained by HE and the pathological changes of liver tissues were observed by light microscope. Results The body weight of rats in each group increased slowly but there was no significant difference (P 〉 0. 05) . The decrease of blood biochemical indexes(TBil, DBil, TBA, AST and ALP) and the improvement of pathological section in the experimental group were obvious( P 〈 0. 05), whilethe decrease of ALT and GGT in the experimental group was not obvious (P 〉 0.05). Conclusion Jinhu Tuihuang Decoction hassignificant effects on removing jaundice, reducing enzyme and repairing hepatocyte injury in rats with cholestatic liver injury induced byacute cholestasis, which may play a role by inhibiting inflammatory reaction and hepatocyte necrosis.
