微小RNA-144-3p及其靶基因NFE2L2在宫颈癌中的表达及临床意义 被引量:6

摘要 目的探讨微小RNA-144-3p(miR-144-3p)及其靶基因NFE2L2在宫颈癌中的表达及临床意义。方法收集新乡医学院第三附属医院2013年1月至2015年12月手术切除的15例宫颈鳞状细胞癌患者的癌组织标本和癌旁组织标本。采用实时荧光定量聚合酶链反应检测宫颈癌组织和癌旁组织中miR-144-3p和NFE2L2 mRNA的相对表达量,采用Western blot检测宫颈癌组织和癌旁组织中NFE2L2蛋白的表达。分析miR-144-3p mRNA相对表达量与患者的年龄、临床分期、细胞分化程度及淋巴结转移的相关性。结果宫颈癌组织和癌旁组织中miRNA-144-3p mRNA的相对表达量分别为0.56±0.20和1.04±0.33,宫颈癌组织中miR-144-3p mRNA的表达量低于癌旁组织(t=-8.38,P<0.05)。miR-144-3p mRNA在宫颈癌组织中的表达与患者年龄无关(P>0.05),与细胞分化程度、临床分期和淋巴结是否转移有关(P<0.05)。NFE2L2 mRNA在宫颈癌组织和癌旁组织中的相对表达量分别为4.41±1.61和0.88±0.10,NFE2L2蛋白在宫颈癌组织和癌旁组织中的相对表达量分别为0.29±0.04和0.15±0.03;NFE2L2mRNA及蛋白在宫颈癌组织中的相对表达量显著高于癌旁组织(t=216.31、-10.97,P<0.05)。结论 miR-144-3p在宫颈癌组织中呈低表达,而其靶基因NFE2L2在宫颈癌组织中呈高表达,miR-144-3p低表达降低了对NFE2L2的抑制作用,进而促进肿瘤的发生和发展。 Objective To study the expression and clinical significance of microRNA-144-3 p(miR-144-3 p) and its target gene NFE2L2 in cervical cancer.Methods Fifteen specimens of squamous cell carcinoma of the cervix and para cancerous tissue were collected in the Third Affiliated Hospital of Xinxiang Medical University from January 2013 to December2015.The expression of miR-144-3 p and NFE2L2 mRNA in cervical cancer tissues and adjacent tissues was detected by realtime polymerase chain reaction;the protein expression of NFE2L2 in cervical cancer tissues and adjacent tissues was detected by Western blot.The correlation between the expression of miR-144-3 p mRNA and age,cellular differentiation,clinical stage and lymph node metastasis was analyzed.Results The relative expression of miR-144-3 p mRNA in cervical cancer tissue and paracancerous tissues was 0.56 ± 0.20 and 1.04 ± 0.33 respectively,the relative expression of miR-144-3 p mRNA in cervical cancer tissue was significantly lower than that in the paracancerous tissues(t=-8.38,P〈0.05).The expression of miR-144-3 p mRNA in cervical cancer was not related to the age of the patients(P〈0.05),which was related to the degree of cell differentiation,clinical stage and lymph node metastasis(P〈0.05).The expression of NFE2L2 mRNA in cervical cancer and paracancerous tissues was 4.41±1.61 and 0.88±0.10,respectively;the expression of NFE2L2 protein in cervical cancer and paracancerous tissues was 0.29 ± 0.04 and 0.15±0.03,respectively.The expression of NFE2L2 mRNA and protein in cervical cancer tissue was significantly higher than that in the paracancerous tissues(t=216.31,-10.97;P〈0.05).Conclusion The expression of miR-144-3 p in cervical cancer tissues is low,and the expression of its target gene NFE2L2 in cervical cancer tissues is high.This study result show that the low expression of miR-144-3 p may be reduce the inhibitory effect on NFE2L2 and promote the occurrence and development of the tumor.
