摘要
为了研究草莓蔗糖转运基因FaSUT5对"申阳"草莓果实糖分积累的影响,提取草莓果实总RNA并反转录为c DNA,并根据GDR数据库中已登录的栽培草莓FaSUT5的全长序列设计引物,对注射了过表达载体35s:SUT5和空载体农杆菌菌株的草莓果实进行了定量分析。研究结果表明:与注射空载体相比,注射了过表达载体的草莓果实中FaSUT5基因的表达量明显提高,蔗糖含量明显提高,而柠檬酸和苹果酸的含量显著下降。
In order to study the influence of strawberry sucrose transporter gene FaSUT5 on the sugar accumulation in fruits of strawberry variety "Shenyang",we extracted the total RNA from strawberry fruits and reversely transcribed it to c DNA,designed specific primers for PCR cloning according to the full-length nucleotide sequence of cultivated strawberry gene FaSUT5 registered in GDR database,and carried out a quantitative analysis of strawberry fruits injected by Agrobacterium strain GV1301 with over-expression vector 35 s: SUT5 or empty vector. The research results indicated that: in the strawberry fruits injected by over-expression vector,the expression quantity of the gene FaSUT5 was evidently increased,and the sucrose content was obviously enhanced,while the contents of citric acid and malic acid were reduced significantly as compared with those in the strawberry fruits injected by empty vector.
