摘要
对一株南极真菌进行菌种鉴定,并初步对其生长条件进行优化。采用形态学观察结合ITS rDNA基因序列鉴定菌株,将扩增片段测序并提交到美国国立生物技术信息中心网站进行BLAST序列比对,分析其序列并进行分子鉴定;通过测量不同温度、不同pH、不同无机盐离子在600nm处的吸光值,初步考察菌株的生长条件。该菌株在斜面培养基上形成黄色至橙黄色光滑菌落,具有多形态细胞;在不同温度,不同pH,不同无机盐离子的培养条件下,菌株的生长状态不同。该菌株为出芽短梗霉菌;菌株在30℃下生长较好,起始pH 5~9时,菌株生长情况差别不大,过多的无机盐离子不利于菌株生长,且易使液体培养基沉淀。
A strain of Antarctic fungus was identified by morphological observation and by ITS rDNA gene sequence and growth conditions were optimized by OD value at 600 nm at different temperature,pH and inorganic salts.The amplified fragments were sequenced and submitted to the NCBI site for Blast sequence alignment for molecular identification.The OD value of the strain was different under different temperature,pH and inorganic salt ions.The yeast strain was identified as Aureobasidium pullulans and had good growth at 30 ℃,without significant difference in growth at pH range of 5—9.Too much inorganic salt was not good for growth of the strains,and led to easy precipitation in liquid medium.
