荞麦属植物RLKs基因序列分析与亲缘关系研究 被引量:3

摘要 从8个荞麦野生种中选取23份荞麦种质资源提取DNA,并根据类蛋白激酶基因(RLKs unigene)序列设计特征引物,进行PCR扩增,对获得的扩增片段进行基因测序与序列分析。结果发现:23份荞麦种质间RLKs基因片段的多态位点数占21.53%,11份野苦荞间RLKs基因片段多态位点数占3.42%,4份野甜荞间多态位点数占1.66%,说明荞麦属植物种内RLKs基因序列高度保守;对扩增序列进行遗传距离分析发现,左贡野甜荞与野甜荞种间遗传距离最小,野甜荞与金荞麦种间遗传距离最大;聚类分析发现,野甜荞与左贡野荞亲缘关系近,与毛野荞和大野荞次之,野苦荞与金荞麦、硬枝野荞和细柄野荞亲缘关系较近。 In the study,23 germplasm resources from 8 wild species were used for DNA extraction. The specific primers were designed based on the high conservative sequence of Receptor-Like Kinases( RLKs) gene for PCR amplification,gene sequencing and analysis. The results showed that 21. 53% of polymorphic sites 23 germplasm resources of genus Fagopyrum were found,while only 3. 42% of polymorphic sites among 11 germplasm resources of F. tataricum and 1. 66% of polymorphic sites among 4 germplasm resources of F. esculentum were found,indicating that RLKs gene sequence is highly conserved in intra-specie of genus Fagopyrum. The minimum value between F. zuogongense and F. esculentum,and the maximum value between F. zuogongense and F. cymosum were found by genetic distance analysis. Hierarchical cluster analysis indicated that the genetic relationship of F. esculentum was close to F. zuogongense and further grouped with F. megaspartanium and F. pilus,while F. tataricum was close to F. cymosum,F. urophyllum and F. gracilipes.
