变应性鼻炎小鼠外周血中滤泡辅助性T细胞的分析 被引量:2

The Analysis of TFH Cells in Mice Model with Allergic Rhinitis

摘要 目的探讨滤泡辅助性T(TFH)细胞在变应性鼻炎小鼠外周血中的数量变化和意义。方法通过卵清蛋白(OVA)及氢氧化铝致敏BALB/c小鼠建立变应性鼻炎模型;TFH细胞用抗小鼠CD4-FITC和抗小鼠CXCR5-PE染色,流式细胞仪计数得到CD4+CXCR5+在小鼠外周血的比例;酶联免疫吸附试验(ELISA)测定血清总Ig E的水平。结果与对照组相比,实验组小鼠鼻腔黏膜下层嗜酸性粒细胞明显增多;实验组OVA特异性Ig E(OVA-s Ig E)浓度(50.44±1.40)pg/m L明显高于对照组(43.47±2.58)pg/m L(P<0.05);实验组外周血CD4+CXCR5+的百分比(13.94±2.77)%明显高于对照组(6.25±1.19)%(P<0.05)。Pearson相关分析显示外周血TFH细胞的数量与OVA-s Ig E浓度呈正相关(r=0.954,P=0.002)。结论变应性鼻炎小鼠外周血中TFH细胞数量增高,参与疾病发生的免疫反应过程,且与Ig E升高的抗体类别转换反应紧密相关。 Objective To investigate the changes and significance of peripheral blood TFH cell in mice with allergic rhinitis. Methods BALB/c mice were sensitized by using OVA and aluminum hydroxide to build the allergic rhinitis model.TFH cells were stained with the anti-mouse CD4 FITC and anti-mouse CXCR5 PE to acquire the quantification of CD4+CXCR5+in blood by flow cytometry.OVA-s Ig E in the serum were measured by enzyme-linked immunosorbent assay( ELISA).Results In the experiment group,there were more eosinophils infiltration in submucosa.The concentration of OVA-specific Ig E increased from( 43.47±2.58) pg/m L to( 50.44±1.40) pg/m L.The percentage of CD4+CXCR5+in blood was up from( 6.25±1. 19) % to( 13.94±2.77) %. There is significantly positive correlation between quantification of circulating TFH and OVA-specific Ig E( r = 0.954,P= 0.002).Conclusion TFH was engaged to the process of allergic immune response and might play an important role in the morbidity of allergic rhinitis.
