ADCY2基因单核苷酸多态性与持续性变应性鼻炎的关联研究 被引量:1

Association study of ADCY2 polymorphisms with persistent allergic rhinitis in a Chinese Han population

摘要 目的探讨腺苷酸环化酶2(ADCY2)基因单核苷酸多态性(SNPs)与苏皖地区汉族人群由尘螨致敏的持续性变应性鼻炎(PER)的遗传相关性。方法采用病例-对照研究,共纳入571例尘螨致敏的PER患者和677例健康对照者。采集所有受试者外周静脉血进行血清总Ig E和特异性Ig E测定,同时EDTA存储管采集抗凝血以提取DNA。预测、筛选ADCY2基因内含子区5个SNPs位点,进行基因分型。结果校正年龄、性别后,ADCY2基因rs1864071位点的基因型频数在病例组和对照组的差异有统计学意义,且该位点在病例组和对照组中的相加模型(OR=1.226,95%CI=1.045-1.439;P=0.013)及显性模型(OR=1.343,95%CI=1.039-1.736;P=0.024)发生PER的风险明显增加。结论在苏皖汉族人群中,ADCY2基因rs1864071位点与PER显著关联,该基因可能影响尘螨致敏PER的遗传易感性。 Objective To explore the association between single nucleotide polymorphisms(SNPs) of adenylate cyclase 2(ADCY2)and persistent allergic rhinitis(PER) due to house dust mite in a Chinese Han population. Methods A case-control study was adapted as 571 patients with mite-sensitized PER and 677 healthy controls were collected. Peripheral venous blood of both groups was drawn to measure serum total Ig E and allergen-specific Ig E, and to extract genome DNA from antithrombin in EDTA containing receptacles. Five SNPs at the introns of ADCY2 gene was selected for genotyping. Results SNP rs1864071 was significantly associated with mite-sensitized PER after adjusted for covariates including age and gender, and patients with addictive model(OR=1.226, 95%CI=1.045-1.439; P=0.013) and dominant model(OR=1.343, 95%CI=1.039-1.736; P=0.024) had an increased risk of PER. Conclusion We observed that there was a significant association of rs1864071 locus with PER in a case-control study, and the ADCY2 gene may contribute to susceptibility of mite-sensitized PER in this Chinese Han population.
