TRPC3在上皮性卵巢癌中的表达及其与ERs的相关性和预后研究

The prognostic significance of TRPC3 expression in epithelial ovarian cancer and its correlation with ERs

目的:探讨瞬时受体电位通道3(TRPC3)在上皮性卵巢癌(EOC)中的表达及其与雌激素受体(ERα、β)的相关性和预后作用。方法:免疫组化法检测TRPC3在91例EOC和10例交界性卵巢肿瘤及22例良性卵巢肿瘤中的表达水平,分析不同卵巢组织中TRPC3的表达差异,TRPC3与ERα、β在EOC中表达的相关性及其临床意义。结果:EOC组织中TRPC3阳性表达率高于良性卵巢肿瘤组织,差异有统计学意义(P<0.05)。Spearman秩相关分析显示,EOC中TRPC3与ERβ表达呈正相关(r=0.233,P=0.026),与ERα表达无相关性(r=0.059,P=0.576)。TRPC3在较晚FIGO分期及高级别癌中呈高表达,差异有统计学意义(P<0.05)。Cox比例风险回归分析提示,单因素分析中TRPC3高表达与患者较短的总生存期(OS)有关(HR=1.97,P=0.025),与无进展生存期(PFS)无关;ERβ高表达在单因素及多因素分析中均与较短PFS有关(HR=2.35,P=0.009;HR=2.26,P=0.014);单因素分析中TRPC3及ERβ均为低表达者与较长OS及PFS均有关(HR=0.50,P=0.023;HR=0.53,P=0.021)。结论:TRPC3及ERβ可能是EOC患者预后不良的预测因子;两者联合可能较单一指标预测EOC患者预后更准确、全面。

Objective： To investigate the expression of transient receptor potential channel 3（ TRPC3） in epithelial ovarian cancer（ EOC） and its correlation with estrogen receptorα、β（ ERα、ERβ） and their prognostic role.Methods： Immunohistochemical method was used to detect the expressions of TRPC3 in 91 cases of EOC,10 cases of borderline ovarian tumors and 22 cases of benign ovarian tumors.To analyze the difference of TRPC3 expression in different ovarian tissues and its correlation with ERα、β and their prognostic value.Results： The positive expression rate of TRPC3 in EOC was higher than that in benign ovarian tumor tissue（ P〈0.05）.Spearman rank correlation analysis showed that there was a positive correlation between TRPC3 and ERβ expression in EOC（ r = 0.233,P = 0.026）,while there was no correlation between TRPC3 and ERα expression（ r = 0.059,P = 0.576）. The expression of TRPC3 was higher in late FIGO stage and higher grade carcinoma,and the difference was statistically significant（ P〈0.05）.The Cox proportional risk regression analysis showed that the high expression of TRPC3 in univariate analysis was related to a shorter overall survival（ OS） of the patients（ HR = 1.97,P = 0.025）,but not to the progressive-free Survival（ PFS）.The high expression of ERβ wasassociated with worse PFS in both univariate and multivariate analysis（ HR = 2.35,P = 0.009;HR = 2.26,P = 0.014）.In univariate analysis,low expression of both TRPC3 and ERβ was associated with longer OS and PFS（ HR = 0.50,P = 0.023; HR = 0.53,P = 0.021）.Conclusions： TRPC3 and ERβ may be the predictors of poor prognosis in patients with EOC and the combination of the two may be more accurate and comprehensive in predicting prognosis than a single indicator.
