促性腺激素释放激素激动剂预防紫杉醇所致小鼠卵巢功能损害的研究 被引量:2

摘要 目的:探讨促性腺激素释放激素激动剂(GnRHa)对紫杉醇(PTX)导致小鼠卵巢功能损害的保护作用。方法:将雌性KM小鼠40只随机分为PTX+Leuprorelin组、PTX组、Leuprorelin组、NS组。用药期间每天观察小鼠的一般状态、动情周期,用药结束后监测小鼠性激素(FSH、E_2、AMH)水平、卵巢湿重、形态结构和各级卵泡数变化。结果:PTX组与NS组比较,小鼠体重明显减轻(P<0.05),卵巢湿重明显下降(P<0.05);PTX组各级卵泡数及卵泡总数显著减少,Leuprorelin组次级卵泡、窦状卵泡数减少,原始、初级卵泡数及总卵泡数增加,PTX+Leuprorelin组原始卵泡比例明显增加,未见明显纤维化及停止发育现象。结论:提前应用GnRHa能减少化疗过程中初级卵泡的丢失,有效预防紫杉醇对小鼠卵巢储备功能的损害,保护卵巢储备功能。 Objective: To explore the effect of gonadotropin-releasing hormone agonist( GnRHa,Leuprorelin) against paclitaxel(PTX) induced gonadotoxicity in mice.Methods: Forty femal KM mice were randomly divided into four groups: PTX+Leuprorelin group,PTX group,Leuprorelin group and NS group. We measured body weight,estrous cycle daily during treatment,and FSH,E2,AMH levels in serum,ovarian weight,histopathology,the number of follicles after treatment,then compared those indicators among the four groups.Results: The body weight of PTX group was significantly lower than NS group( P〈0.05),and the wet weight of ovary significantly decreased( P〈0.05).The follicle number and total number of follicles in PTX group were significantly decreased under microscope. The number of primordial follicles and the total number of follicles increased,the proportion of primordial follicles in PTX +Leuprorelin group increased significantly,no obvious fibrosis and the phenomenon of arrest were observed.It could be seen that Leuprorelin significantly improve the damage of PTX to ovarian tissue and effectively protect the ovarian reserve Features.Conclusion: Early application of gonadotropin-releasing hormone agonist( GnRHa) can effectively prevent paclitaxel on ovarian reserve function in mice damage,reduce the loss of primary follicles during chemotherapy.
