摘要
目的:综合评价3个单核苷酸多态性位点(SNPs)rs12478601、rs13405728、rs13429458与多囊卵巢综合征(PCOS)发病的相关性。方法:计算机检索Pub Med、CBM、CNKI、VIP、万方数据库2017年11月以前发表的3个单核苷酸多态性位点(SNPs)rs12478601、rs13405728、rs13429458与PCOS发病相关性的病例-对照研究。评价文献质量,采用Rev Man5.3软件进行meta分析。结果:共纳入10组独立病例-对照研究。Meta结果显示,亚洲人群纯合子模型中病例组的SNP-458(A)(OR=0.5,95%CI为0.42~0.61,P<0.00001)、SNP-728(A)(OR=0.49,95%CI为0.38~0.62,P<0.00001)、SNP-601(C)(OR=0.53,95%CI为0.46~0.62,P<0.00001)明显高于对照组,差异有统计学意义。亚洲人群显性模型分析结果示,病例组SNP-458(A)(OR=0.69,95%CI为0.64~0.74,P<0.00001)、SNP-728(A)(OR=0.66,95%CI为0.60~0.71,P<0.00001)、SNP-601(C)(OR=0.69,95%CI为0.60~0.79,P<0.00001)高于对照组,差异有统计学意义。欧美人群纯合子模型中,病例组与对照组比较SNP-458(A)(OR=0.58,95%CI为0.24~1.41,P=0.23)、SNP-728(A)(OR=0.99,95%CI为0.69~1.43,P=0.96)、SNP-601(C)(OR=0.91,95%CI为0.76,1.09,P=0.30)差异无统计学意义。结论:亚洲人群中SNP-728的等位基因A、SNP-458的等位基因A和SNP-601的等位基因C可能增加PCOS的患病风险,欧美人群中3个位点可能与其致病性无关。
Objective: To evaluate the association between PCOS susceptibility and three reported single nucleotide polymorphisms( SNPs),namely,rs12478601,rs13405728 and rs13429458.Method: Pub Med,CBM,CNKI,VIP and Wanfang databases were searched to obtain the case-control results till Nov.2017.We evaluated the quality of the results by using RevMan5.3 software. Results: A total of 10 independent case-control studies were retrieved from these databases,and all of the meta-analysis were performed with both the homozygous and dominant models.The results showed that all of the three SNPs were significantly higher than the control group in Asian population with the homozygous model: SNP-458( A)( OR = 0.5,95%CI0.42 ~ 0.61,P〈0. 00001),SNP-728( A)( OR = 0. 49,95% CI 0. 38 ~ 0. 62,P〈0.00001) and SNP-601( C)( OR = 0.53,95% CI 0.46 ~ 0.62,P〈0.00001). Furthermore,the dominant model results also obtain the similar results: SNP-458( A)( OR = 0. 69,95% CI 0. 64 ~ 0. 74,P〈0.00001),SNP-728( A)( OR = 0.66,95%CI 0.60 ~ 0.71,P〈0.00001),SNP-601( C)( OR = 0.69,95%CI 0.60 ~ 0. 79,P〈0. 00001). While for the Euro-American populations,there were no significant differences between the case group and the control group: SNP-458( A) was compared( OR = 0.58,95%CI 0.24 ~ 1.41,P = 0. 23),SNP-728( A)( OR = 0.99,95% CI 0. 69 ~ 1.43,P = 0.96),SNP-601( C)( OR = 0.91,95% CI 0.76 ~ 1.09,P = 0.30). Conclusion: In Asian,SNP-728( A),SNP-458( A) and SNP-601( C) may increase the risk of PCOS,and the three SNPs in Euro-American populations might be not related to the susceptibility of PCOS.
