摘要
目的探讨MRP1、BCRP、BRCA1在上皮性卵巢癌中的表达情况及其意义。方法 47例上皮性卵巢癌患者卵巢组织标本,根据耐药情况分为耐药组(22例)和敏感组(25例),另选正常卵巢组织标本作为对照组(20例)。采用实时荧光定量PCR技术对上皮性卵巢癌组织和正常卵巢组织中MRP1、BCRP、BRCA1基因的表达情况进行检测。结果 MRP1在上皮性卵巢癌组织中的表达量和表达率均高于正常卵巢组织;BCRP在上皮性卵巢癌组织表达量和表达率均低于正常卵巢组织(P<0.05);BRCA1在上皮性卵巢组织中的表达量和表达率与正常卵巢组织比较差异无统计学意义(P>0.05)。耐药组MRP1、BRCA1表达量和表达率均高于敏感组;BCRP表达量和表达率均低于敏感组(P<0.05)。结论 MRP1和BCRP在上皮性卵巢癌组织中的表达量会显著增加,BRCA1表达量变化不大。MRP1和BCRP表达量的增加可能与化疗耐药有关,影响患者疗效。
Objective To explore the expressions of MRP1,BCRP and BRCA1 in epithelial ovarian carcinoma and its association with chemotherapy drug resistance.Methods Cancer tissue samples collected from 47 patients with epithelial ovarian carcinoma were divided into drug-resistant group( n = 22) and non-resistance group( n = 25) according to resistance status.At the same time,20 normal ovarian tissue samples were taken as control group.The gene expressions of MRP1,BCRP and BRCA1 were detected by using real-time PCR technology.Results The expression quantity and rate of MRP1 in the cancer tissue were significant higher than that in control group( P〈0. 05). The expression quantity and rate of BCRP in the cancer tissues were significant lower than that in control group( P〈0. 05).There was no significant difference in expression quantity and rate of BRCA1 between the cancer tissues and the control group( P〈0. 05).The expression quantity and rate of MRP1 and BRCA1 in the resistance group were significantly higher than those in the non-resistance group( P〈0. 05).On the other hand,the BCRP expression quantity and rate in the resistance group were significantly lower than that in the non-resistance group( P〈0. 05).Conclusion The MRP1 and BCRP expressions in epithelial ovarian cancer are increased significantly while the BRCA1 expression does not change significantly. The increase in expression of MRP1 and BCRP could result in drug resistance and is bad for curative effect.
