摘要
目的建立同时测定益心通脉颗粒中丹参酮Ⅰ、隐丹参酮和丹参酮ⅡA含量的HPLC法。方法采用Luna 5μm C18(2)(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-水(67∶33);检测波长:270nm;流速:1.0mL·min^(-1);柱温:40℃。结果丹参酮Ⅰ、隐丹参酮和丹参酮ⅡA的线性范围分别为0.042~1.260(r=1.000),0.036~1.073(r=1.000)和0.032~0.948μg(r=1.000),平均回收率分别为97.4%(RSD=1.2%),99.5%(RSD=1.6%)和97.6%(RSD=0.8%)。结论经方法学验证,本方法合理可行、重复性好,可用于益心通脉颗粒中丹参酮Ⅰ、隐丹参酮和丹参酮ⅡA的含量测定,为益心通脉颗粒的质量控制提供依据。
Objective To establish an HPLC method for the determination of tanshinoneⅠ,cryptotanshinone and tanshinoneⅡA in Yixintongmai Granules.Methods The HPLC experiments were performed on a Luna 5μm C18(2)column(250 mm×4.6 mm,5μm)with 270 nm as the detection wavelength,and methanol-water(67∶33)as the mobile phase.The column temperature was 40℃,and the flow rate was 1.0 mL·min^(-1).Results The linear ranges of tanshinoneⅠ,cryptotanshinone and tanshinoneⅡA were 0.042-1.260(r=1.000),0.036-1.073(r=1.000)and 0.032-0.948μg(r=1.000)respectively.The average recoveries were97.4%(RSD=1.2%),99.5%(RSD=1.6%)and 97.6%(RSD=0.8%)respectively.Conclusion The improved methods are reasonable,feasible and reproducible,which could be used for the quality control of Yixintongmai Granules.
