摘要
目的研究β-七叶皂苷钠在大鼠高压电损伤早期的抗渗作用。方法将100只SD大鼠随机分为β-七叶皂苷钠组和对照组,分别制成下肢高压电损伤模型后即刻静脉注射β-七叶皂苷钠(β-七叶皂苷钠组)及等量生理盐水(对照组)。各组分别按电烧伤后1、6、24、48、72h分为5小组,每小组10只,大体观察后取创周组织参照改良伊文思蓝渗出法行血管通透性测定及组织含水量测定。结果β-七叶皂苷钠组1、6、24、48h和72h 5个时相点伊文思蓝(EB)浓度分别为(293.15±64.45)μg/g、(239.37±41.47)μg/g、(259.49±33.74)μg/g、(243.62±32.78)μg/g、(226.25±29.69)μg/g,对照组相应EB浓度为(402.10±43.22)μg/g、(282.45±28.05)μg/g、(322.65±29.01)μg/g、(300.66±41.15)μg/g、(273.01±39.43)μg/g,β-七叶皂苷钠组与对照组相比差异均有统计学意义,P<0.05或P<0.01。β-七叶皂苷钠组1、6、24、48h和72h 5个时相点组织含水量分别为(73.08±2.04)%、(80.47±2.23)%、(78.36±1.97)%、(77.22±1.15)%、(75.16±3.09)%,对照组相应时间点组织含水量为(76.30±2.60)%、(84.68±1.71)%、(82.18±2.09)%、(80.57±2.27)%、(79.15±2.22)%,与对照组相比,β-七叶皂苷钠组各时相点差异均有统计学意义,P<0.05或P<0.01。与对照组相比,β-七叶皂苷钠组血管通透性及组织含水量均降低。结论β-七叶皂苷钠对高压电损伤大鼠具有抗渗作用。
Objective To investigate the anti-permeability effect of β-aescin on early rats after high voltage electrical injury. Methods One hundred rats were randomly divided into β-aescin group and normal saline group,which were given β-aescin and normal saline,respectively by caudal vein after injury. Experimental animal models of electrical injury were made for β-aescin group and normal saline group. Ten rats of each group were killed at 1,6,24,48 and 72 hours after injury. Tissue samples around the wound were harvested,capillary permeability and water content were measured to observe the changes of the microvascular structure. Results The level of Evans blue in the β-aescin group at 1,6,24,48 and 72 hours after injury was( 293. 15 ± 64. 45) μg/g,( 239. 37 ± 41. 47) μg/g,( 259. 49 ± 33. 74) μg/g,( 243. 62 ± 32. 78) μg/g and( 226. 25 ± 29. 69) μg/g,respectively,which was( 402. 10 ±43. 22) μg/g,( 282. 45 ± 28. 05) μg/g,( 322. 65 ± 29. 01) μg/g,( 300. 66 ± 41. 15) μg/g and( 273. 01 ± 39. 43)μg/g in the normal saline group,respectively. There was statistically significant difference between the two groups( P〈0. 05 or P〈0. 01). The water content in the β-aescin group at 1,6,24,48 and 72 hours after injury was73. 08% ± 2. 04%,80. 47% ± 2. 23%,78. 36% ± 1. 97%,77. 22% ± 1. 15% and 75. 16% ± 3. 09%,respectively,which was 76. 30% ± 2. 60%,84. 68% ± 1. 71%,82. 18% ± 2. 09%,80. 57% ± 2. 27% and 79. 15% ± 2. 22%,respectively in the normal saline group. There was statistically significant difference between the two groups( P〈0. 05 or P〈0. 01). Compared with the normal saline group,there were lower capillary permeability and water content in the β-aescin group. Conclusion β-aescin has anti-permeability effect on rats after high voltage electrical injury.
