水苏碱对急性心肌缺血小鼠的保护作用 被引量:3

摘要 目的观察水苏碱对急性心肌缺血损伤小鼠的防治作用及其可能机制。方法小鼠50只随机分为5组(n=10):正常对照组、模型对照组、水苏碱低、中、高剂量组(5 mg/kg、10 mg/kg、20 mg/kg)。采用皮下注射异丙肾上腺素制备心肌缺血小鼠模型,不同剂量水苏碱于造模前腹腔注射给药,连续7 d。测定小鼠血清乳酸脱氢酶(LDH)、肌酸激酶(CK)活性,并测定心肌组织Na^+-K^+-ATPase,Ca^(2+)-Mg^(2+)-ATPase活性及髓过氧化物酶(MPO)活性,HE染色观察心肌组织的病理形态学变化。结果水苏碱可降低血清LDH、CK活性和心肌MPO活性,提高心肌Na^+-K^+-ATPase,Ca^(2+)-Mg^(2+)-ATPase活性(P<0.05)。结论水苏碱对心肌缺血小鼠有保护作用,其机制与改善细胞能量代谢及抑制炎症反应有关。 Objective: To investigate the protective elfect of Stachydrine on acute myocardial ischemia and its mechanism in mice. Methods: Mice were randomly divided into five groups( n = 10) : normal control group,model group and Stachvdrine low-dose,medium-dose and high-dose groups( 5 mg/kg,10 mg/kgand 20 mg/kg). The mice were subcutaneously injected with isoprenaline to induce myocardial ischemia model. The three doses of Stachvdrine were administered through intraperitoneal injection once daily for seven days. The activities of Na^+-K^+-ATPase,Ca^(2+)-Mg^(2+)-ATPase and MPO in myocardial tissue were detected. Hematoxylin-eosin staining was explored to obersve myocardial ischemic injury.Results: Stachvdrine significantly reduced the LDH and CK activity in serum. Stachvdrine reduced MPO activity and increased Na^+-K^+-ATPase,Ca^(2+)-Mg^(2+)-ATPase activities in myocardial tissue. Conclusion: Stachvdrine preconditioning exerts the marked protective effect on acute myocardial ischemia injury in mice,which is related to regulate energy metabolism and anti-inflammation effect.
