逆境诱导型启动子Mvnhx1缺失片段植物表达载体构建

摘要 Mvnhx1启动子是一种与盐胁迫和干旱胁迫相关的逆境诱导型启动子。为了更好地研究顺式作用元件对Mvnhx1启动子功能特点的影响,研究根据启动子序列上与耐盐和耐旱相关的顺式作用元件分布位置不同设计引物,利用5'端缺失技术获得11个不同长度的Mvnhx1启动子缺失片段,分别构建不同缺失片段与报告基因GUS融合的植物表达载体。PCR扩增结果显示,成功克隆获得11个长度分别为1 135、1 027、974、967、858、727、726、564、366、352和210 bp的片段。酶切鉴定显示,成功构建了由Mvnhx1启动子11个不同长度缺失片段调控的带GUS基因的植物表达载体。 The Mvnhx1 promoter is an adversity-inducible promoter associated with salt stress and drought stress. In order to better study the effect of cis-acting elements on the functional characteristics of Mvnhx1 promoter, the primers were designed according to the distribution position of cis-acting elements related to salt tolerance and drought tolerance in the promoter sequence, and the results were obtained by 5' Eleven different length of the Mvnhx1 promoter fragment were fragmented to construct a plant expression vector with different deletion fragments and the reporter gene GUS. The results of PCR amplification showed that 11 fragments were 1 135 bp, 1 027 bp, 974 bp, 967 bp, 858 bp, 727 bp, 726 bp, 564 bp, 366 bp, 352 bp and 210 bp, respectively. The results of restriction enzyme digestion showed that the plant expression vector of GUS gene regulated by 11 different deletions of Mvnhx1 promoter was successfully constructed.
