罗格列酮对非酒精性脂肪肝大鼠脂联素及胰岛素抵抗的影响 被引量:3

摘要 目的观察罗格列酮对非酒精性脂肪肝(NAFLD)大鼠脂联素及胰岛素抵抗的影响。方法 10只大鼠为对照组,余30只为造模组,给予高脂饮食,第8周时,随机处死对照组和造模组各1只大鼠,常规肝切片行HE染色,以肝细胞明显脂肪变性确定制备模型成功。将造模组随机分为高脂组和罗格列酮组,前者继续以高脂喂养;后者自第9周开始,每天以5mg·kg^(-1)·d^(-1)罗格列酮盐溶液灌胃,共8w。各组大鼠在实验第18周末测量体质量,麻醉后计算肝脏指数,测定血清中丙氨酸氨基转移酶(ALT)、胆固醇(TC)和甘油三酯(TG)含量;脂联素采用酶联免疫吸附(ELISA)法检测。结果高脂组大鼠体质量与肝脏指数均明显增高(P<0.05)。罗格列酮组的肝脏指数较高脂组比明显降低;与对照组比较,肝脏指数有所升高(P<0.05)。对照组、高脂组脂联素水平明显低于对照组,而罗格列酮组脂联素水平明显高于对照组及高脂组(P<0.05)。与对照组比较,高脂组大鼠血清ALT、AG和TC水平明显高于对照组(P<0.05),而罗格列酮组明显低于高脂组(P<0.05)。结论罗格列酮能有效逆转大鼠酒精性脂肪肝,改善胰岛素抵抗。 Objective To observe the effect of rosiglitazone on adiponectin and insulin resistance in non-alcoholic fatty live disease(NAFLD)rats.Methods Ten rats were used as normal control group,and other30 rats fed with high-fat diet were set up as the model group.Conventional HE staining was used to confirm successful establishment of the model of hepatocellular steatosis from randomly sacrificed semi-model rats.Then the model group was randomly divided into high fat group and rosiglitazone group.Starting from the 9 th week,the rats were orally gavaged with 5 mg/kg rosiglitazone daily for 8 weeks.The rats in the control group and the rats in the high fat group were given the same volume of normal saline at the same time.Body weight was measured at the end of the 18 th week in each group,liver index(liver index=wet weight of liver/body weight)was calculated after anesthesia,serum alanine aminotransferase(ALT),cholesterol(TC)and triglyceride(TG)content were measured.Adiponectin was detected by enzymelinked immunosorbent assay(ELISA).Results The body weight and liver index of rats in high fat diet group were significantly increased(P〈0.05),while there was no significant difference between the control group and rosiglitazone group in body weight and liver index(P〈0.05).Compared with the control group,the liver index of the rosiglitazone group was significantly lower than that of the high lipid group with statistically significant(P〈0.05).The levels of adiponectin in the control group,high fat group and rosiglitazone group were significantly different(P〈0.05).The levels of adiponectin in the control group,high fat group and rosiglitazone group were(79.2±13.1),(69.4±13.9)and(89.4±14.1)μg/mL respectively.Adiponectin levels of rosiglitazone group were lower than the control group,adiponectin levels were higher than the control group,and the high-fat group,the difference was statistically significant(P〈0.05).The levels of serum ALT,AG and TC in the high fat diet group were significantly higher than those in the control group.Serum levels of ALT,AG and TC in rosiglitazone group were significantly lower than those in high fat diet group.There were no significant differences in serum indexes between rosiglitazone group and control group(P〈0.05).Conclusion Rosiglitazone can effectively reverse alcoholic fatty liver in rats and improve insulin resistance.
