摘要
以进口荷兰黄水仙、多花水仙和中国水仙为研究对象,进行DNA提取、PCR扩增和数据分析,来评估mat K基因、rbc L基因、trn H-psb A片段和ITS片段4个候选DNA条形码的鉴定能力.建树法分析结果显示:叶绿体基因片段具有鉴定黄水仙品种的能力,但中国水仙与黄水仙聚为一支;核基因片段ITS序列可以准确区分出中国水仙和黄水仙,但在区分黄水仙品种时支持率较低;4种基因片段的组合条形码具有较强的鉴定能力,能准确鉴定出中国水仙和黄水仙,也能识别出不同黄水仙品种间的亲缘关系.距离分析法的结果显示,ITS+mat K、ITS+trn H-psb A、ITS+rbc L和ITS+mat K+trn H-psb A+rbc L 4种组合条形码鉴定的成功率较高,适用于黄水仙品种的鉴定.
DNA extraction,PCR amplification and data analysis were carried out to evaluate the identification ability of 4 candidate DNA barcodes of genes mat K,rbc L,and fragments trn H-psb A,ITS from Chinese Narcissus and foreign species. The results showed that chloroplast gene fragments had the ability to identify varieties of Narcissus,but Chinese Narcissus and Dutch Narcissus are clustered into one. Nuclear gene fragment ITS sequence could accurately distinguish Chinese Narcissus and Dutch Narcissus,but it had lower support rate in distinguishing Narcissus species. The combination barcodes of the 4 gene fragments had strong identification ability. It could identify Chinese Narcissus and Dutch Narcissus,and could identify the relationship between different varieties of Dutch Narcissus. The results of distance analysis showed that the success rate of the 4 combinations of ITS+mat K,ITS+trn H-psb A,ITS+rbc L and ITS+mat K+trn H-psb A+rbc L was higher,which is suitable for the identification of the variety of the Dutch Narcissus.
