微小核糖核酸-146a调节CXC型趋化因子4对甲状腺癌细胞侵袭能力的影响 被引量:3

摘要 目的探讨微小核糖核酸(miR)-146a通过调节CXC型趋化因子4(CXCR4)对甲状腺癌SW579细胞侵袭能力的影响,了解miR-146a参与甲状腺癌发生、发展的分子机制。方法设计miR-146a mimics对SW579细胞进行转染,采用实时定量PCR检测各组miR-146a和CXCR4 mRNA的表达,利用Western blot检测各组CXCR4蛋白的表达,采用Transwell检测转染后SW579细胞侵袭能力的变化。结果转染miR-146a mimics后,SW579细胞中CXCR4 mRNA和蛋白表达量较空白对照组和阴性对照组降低(P<0.05);SW579细胞的侵袭能力较空白对照组和阴性对照组显著降低(P<0.05)。结论 MiR-146a可能通过抑制CXCR4的表达来抑制甲状腺癌细胞的侵袭能力,miR-146a可能参与甲状腺癌的发生、发展。 Objective To investigate the effect of micro ribonucleic acid(miR)-146 a by regulating CXC chemokine receptor 4( CXCR4) on the ability of invasion of thyroid cancer SW579 cells. Methods Chemically synthesized miR-146 a mimics were transfected into SW579 cells. The expressions of miR-146 a and CXCR4 mRNA were detected by real-time PCR. The expressions of CXCR4 protein were detected by Western blot. The effect on the ability of invasion was detected by Transwell. Results SW579 cells transfected miR-146 a mimics significantly downregulated the expression of CXCR4 mRNA and protein as compared with the blank control group and the negative control group( P〈0.05). After transfection with miR-146 a mimics,the ability of invasion of SW579 cells was lower than those in the blank control group and the negative control group( P〈0.05). Conclusion MiR-146 a may inhibits the invasion of thyroid cancer cells by regulating CXCR4.
