TCF7L2基因rs290487位点多态性与新疆维吾尔族、汉族2型糖尿病中医基本证候关系分析

Study on correlation between Rs290487 Variant in TCF7L2 gene and type 2 diabetes in Uygur and Han

目的分析TCF7L2基因rs290487位点多态性与新疆维吾尔族、汉族2型糖尿病中医基本证候的相关性,以指导辨证分型。方法选择2012年3月—2014年12月于新疆医科大学附属中医院内分泌科住院治疗的维吾尔族2型糖尿病患者288例、汉族2型糖尿病患者292例作为研究对象。采用多聚酶联反应—限制性片段长度多态性法进行TCF7L2基因rs290487位点多态性分析,同时对2型糖尿病患者的中医证候进行调查分析,再关联分析中医证候与TCF7L2基因rs290487位点多态性的交互作用。结果维吾尔族和汉族2型糖尿病患者中医证型中均以脾虚痰湿证分布最多,其次为气虚血瘀证,其他证型分布最少。不同族别的中医证型分布比较,差异无统计学意义(P>0.05)。TCF7L2基因rs290487位点基因型在维吾尔族和汉族患者各中医证型所占比例比较差异均有统计学意义(χ~2=4.702、4.238,P=0.031、0.042);维吾尔族患者中脾虚痰湿证与气阴两虚证比较差异有统计学意义(χ~2=4.362,P=0.037),汉族患者中气虚血瘀证与其他证型组比较差异有统计学意义(χ~2=6.693,=0.010)。结论维吾尔族和汉族2型糖尿病患者的中医证候分型与TCF7L2基因rs290487多态性位点均有内在关系。

Objective To study the correlation between Rs290487 Variant in TCF7 L2 gene and type 2 diabetes mellitus（ DM） in Uygur and Han. Methods Collected 580 patients with type 2 DM the research objects, including 288 cases of Uygur and 292 cases of Han. Rs290487 TCF7 L2 gene loci polymorphism analysis was genotyped by polymerase reaction restriction fragment length polymorphism method. The correlation analysis of syndrome and the rs290487 TCF7 L2 gene loci polymorphism interaction were performed. Results In Uygur and Han patients with type 2 DM most were phlegm dampness syndromes distribution of pixu（spleen deficient）,followed by vein blood stasis syndrome. The of comparison, there was no statistically significant difference of syndrome type distribution between Uygur and Han patients（ P〈0. 05）. Rs290487 TCF7 L2 gene loci genotypes in Uygur and Han patients of different syndrome type was statistically significant（χ^2=4.702, χ^2=4. 238,P = 0. 031, P = 0. 042）.,There were statistically significant in Uygur patients with phlegm dampness syndromes pixu（ spleen deficient） or qi and Yin deficiency syndrome（χ^2=4. 362, P = 0. 037）. The Han patients with blood stasis showed statistically significant difference with other kind of syndrome（χ^2=6. 693,P = 0. 010）. Conclusion The TCM syndrome classification of Uygur and Han patients with type 2 DM is closely related to the rs290487 polymorphism of TCF7 L2 gene.
