摘要
目的探讨改变胃癌细胞自噬水平对其侵袭转移能力的影响。方法将胃癌SGC-7901细胞分为空白对照组(CON组)、乏氧组(HPX组)及乏氧联合巴弗洛霉素A1(BAF-A1)组(H+B组)。分别采取吖啶橙染色及Western blot检测胃癌细胞自噬水平;Transwell法检测细胞侵袭转移能力;Western blot检测侵袭转移相关蛋白表达情况。结果与CON组相比,HPX组红色荧光量显著增多,Beclin1相对表达量显著增高(P=0.000),侵袭转移细胞数显著增多(P=0.001),E-Cadherin蛋白相对表达量显著降低(P=0.021),MMP-9蛋白相对表达量显著增高(P=0.013);与HPX组相比,H+B组红色荧光量显著减少,Beclin1相对表达量显著降低(P=0.000),侵袭转移细胞数显著减少(P=0.000),ECadherin蛋白相对表达量显著增高(P=0.001),MMP-9蛋白相对表达量显著降低(P=0.001)。结论乏氧状态提高胃癌细胞自噬水平后其侵袭转移能力显著增强,而当其自噬水平被BAF-A1抑制后胃癌细胞的侵袭转移能力显著降低。
Objective To investigate the effect of changing autophagic level on invasive and migratory ability of gastric cancer cells.Methods The SGC-791 cells were divided into three groups,the control group,the hypoxia group and the hypoxia+bafilomycin A1 group.Acridine orange staining method and Western blot were used to detect the cell autophagy.Transwell chamber was used to detect the invasion and migration of gastric cancer cells.Western blot was used to detect the expression of E-Cadherin and MMP-9.Results Compared with the control group,the amount of red fluorescence in the hypoxia group was more,the expression of Beclin 1 was higher,the invasive and migratory cells were more,the expression of E-Cadherin was lower,and the expression of MMP-9 was higher.Compared with the hypoxia group,the amount of red fluorescence in the hypoxia+bafilomycin A1 group was less,the expression of Beclin 1 was lower,the invasive and migratory cells were less,the expression of E-Cadherin was higher,and the expression of MMP-9 was lower.Conclusion When the cells were lack of oxygen,the autophagic level became higher,and it would enhance the invasive and migratory ability.When the autophagy was inhibited by bafilomycin A1,the invasive and migratory ability of gastric cancer cells was suppressed.
