苯基苯并呋喃类化合物的区域选择性合成方法 被引量:1

摘要 苯并呋喃类化合物广泛存在于天然产物中,具有多种生物活性,是重要的医药中间体。为了探索合成此类化合物的有效方法,由一系列酚类与α-溴代苯乙酮在碱性条件下反应得到酚醚中间体,酚醚在多聚磷酸(PPA)的催化作用下实现电环化反应脱水制备苯基苯并呋喃。并研究了PPA存在下3-苯基萘并呋喃转化为2-苯基萘并呋喃的异构化反应动力学,通过控制反应温度,发现这一转化受位阻影响较大。高选择性的合成了一系列2-或3-苯基取代的产物。 Benzofurans are widely spread natural products with various biological activities,which makes them the important intermediates for drug synthesis. In this report,the reaction of a series of phenols with α-bromoacetophenone to afford the phenolic ether intermediates was investigated. And the electro-cyclic condensation reactions of the phenolethers catalyzed by poly phosphoric acid( PPA) could selectively afford 2-or 3-phenyl benzofurans by controlling the reaction temperatures.The isomerization of 3-phenyl naphathofuran to 2-phenyl naphathofuran in the presence of PPA was studied.
