摘要
目的建立并优化竞争法酶联免疫吸附试验(ELISA)对弹性蛋白降解产物(EDP)的检测;观察弹性蛋白和EDP在肝硬化患者中的表达特点。方法回顾性纳入失代偿期肝硬化患者14例,健康对照人群10例,应用竞争法ELISA检测两组人群EDP表达水平,应用商品化ELISA试剂盒检测两组人群弹性蛋白水平。根据建立的剂量-反应曲线和曲线拟合的程度,选择最佳的竞争法ELISA检测EDP的条件。结果成功建立并优化竞争法ELISA检测EDP的条件,标准曲线呈线性拟合,R^2=0.99。通过对弹性蛋白和EDP的检测发现,肝硬化患者弹性蛋白的水平显著高于健康对照组[(32.19±12.17)ng/ml vs.(1.90±0.46)ng/ml,P<0.05)],而EDP的表达显著低于健康对照组[(28.20±1.80)μg/ml vs.(60.80±1.46)μg/ml,P<0.05]。结论弹性蛋白和EDP水平与失代偿期肝硬化相关。
Objective To establish and optimize the method of competitive enzyme-linked immunosorbent assay( ELISA) for elastin derived peptides( EDP),meanwhile to observe the expression of elastin and EDP in patients with cirrhosis. Methods According to the establishment of dose-response curve,the competitive ELISA for EDP were optimized. This is a retrospective study,which were recruited 14 cases of decompensated liver cirrhosis patients and 10 cases in healthy control group. Expression of elastin and EDP were measured using competitive ELISA and commercial ELISA kit respectively. Results The competitive ELISA for EDP were established and optimized successfully. The standard curves were linear fitted and R^2= 0. 99. The expression of elastin in cirrhotic patients was significantly higher than healthy control group [( 32. 19± 12. 17) ng/ml vs( 1. 90 ± 0. 46) ng/ml,P〈0. 05],meanwhile EDP expression was significantly lower than healthy control group [( 28. 20 ±1. 80) μg/ml vs( 60. 80 ± 1. 46) μg/ml,P〈0. 05]. Conclusion These findings suggest that levels of elastin and EDP are associated with the development of decompensated liver cirrhosis.
