摘要
从白桦组织中克隆2个响应Me JA、SA信号诱导的bHLH家族基因,分别命名为BpbHLH2和BpbHLH3,并对其进行生物信息学及表达特性分析。结果表明:2个bHLH基因均具有完整的开放读码框(ORF),BpbHLH2基因ORF长1 050 bp,编码349个氨基酸,BpbHLH3基因ORF长1 044 bp,编码347个氨基酸。两个基因编码的氨基酸序列与胡桃(Juglans regia)bHLH家族蛋白(Gen Bank ID:XP_018817560.1)和(Gen Bank ID:XP_018813766.1)相似度最高,分别达71%和70%。2个基因均包含bHLH、E-box和N-box的保守序列。BpbHLH2、BpbHLH3基因在白桦的根、茎、叶中均有表达,且两个基因均在叶中的表达量最高,BpbHLH2、BpbHLH3分别在根和茎中的表达量较低。生物信息学分析显示,BpbHLH2、BpbHLH3为白桦bHLH家族中的新基因。
Two bHLH family genes from Betula platyphylla Suk. in response to MeJA and SA signaling were cloned, named BpbHLH2 and BpbHLH3 , respectively, and bioinformatics and expression characteristics of two genes were analyzed in this study. The two bHLH genes contain complete ORF (opening reading frame), BpbHLH2 gene ORF was 1050 bp, encoding 349 amino acids, BpbHLH3 gene ORF was 1044 bp, encoding 347 amino acids The amino acid sequence was similar to bHLH family protein of Juglans regia (GenBankID:XP_018817560.1) and (GenBankID:XP_018813766.1), respectively, with a maximum of the amino acid sequence is similarity to bHLH family protein of Juglans regia , up to 71% and 70%. The two genes contain conserved sequences of bHLH, E-box and N-box. BpbHLH2 and BpbHLH3 genes were expressed in roots, stems and leaves of Betula platyphylla Suk.. The two genes expressed the highest in leaves, and the expression of BpbHLH2 and BpbHLH3 in roots and stems was lower respectively. By bioinformatics analysis, BpbHLH2 and BpbHLH3 genes were new bHLH family members from Betula platyphylla Suk.
