摘要
目的探讨胰岛素样生长因子-2(IGF-2)对婴幼儿血管瘤干细胞(Hem SCs)增殖及向脂肪分化的影响。方法搜集3例增生期草莓状婴幼儿血管瘤术后组织,采用胶原酶消化分离后CD133免疫磁珠吸附得到血管瘤干细胞。CCK-8法检测不同浓度IGF-2对Hem SCs增殖的影响,Western blot检测空白对照组及IGF-2组(100 ng/ml)、IGF-2+OSI-906组(IGF-2:100 ng/ml;OSI-906:IGF-1R受体抑制剂1μmol/L)、IGF-2+LY294002组(LY294002:PI3K抑制剂10μmol/L)、LY294002组(10μmol/L)各组细胞中脂肪转化因子C/EBPα、C/EBPβ、PPARγ、adiponectin、p-AKT及total AKT的表达。结果 IGF-2在体外对Hem SCs增殖有明显促进作用(P<0.05);Western blot法显示,与IGF-2+OSI-906组、IGF-2+LY294002组、LY294002组及空白对照组比较,IGF-2组C/EBPα、C/EBPβ、PPARγ、adiponectin表达量增加(P<0.05);与IGF-2+OSI-906组、IGF-2+LY294002组及空白对照组比较,IGF-2组p-AKT表达量增加(P<0.05)。结论 IGF-2可能通过IGF-1R/PI3K/AKT通路影响HemS Cs的脂肪转化过程。
Objective To investigate the effect of insulin-like growth factor-2(IGF-2) on the proliferation and adipogenesis differentiation of hemangioma-derived stem cells.Methods The proliferating infantile hemangiomas tissues resected from three patients were collected.After collagenase digestion and separation,CD133 immunomagnetic beads were adsorbed to obtain hemangioma-derived stem cells.CCK-8 assay of different concentrations of IGF-2 on the proliferation of Hem SCs,cells were cultured in EGM-2/FBS-10% containing IGF-2(100 ng/ml),IGF-2 plus OSI-906(IGF-2:100 ng/ml;OSI-906:1 μmol/L),IGF-2 plus LY294002(IGF-2:100 ng/ml;LY294002:10 μmol/L),LY294002(10 μmol/L) or no treatment,the protein expression of C/EBPα,C/EBPβ,PPARγ,adiponectin,p-AKT and total AKT were detected.Results IGF-2 significantly enhanced Hem SCs proliferation in vitro,the difference was statistically significant(P0.05).Western blot showed that the expression of C/EBPα,C/EBPβ,PPARγ,adiponectin and p-AKT in IGF-2 group were increased compared with the other groups.Conclusion IGF-2 may affect the adipogenesis differentiation of Hem SCs through the IGF-1 R/PI3 K/AKT pathway.
