BTG1在胃癌中的表达及其临床意义

Expression of BTG1 in gastric cancer and its clinical significance

目的分析B细胞异位基因1(BTG1)在胃癌中的表达及其潜在临床意义。方法收集胃癌组织和相对应癌旁组织标本46例及健康胃黏膜组织标本31例,利用荧光定量PCR技术和免疫组织化学技术分别检测各标本中BTG1的mRNA和蛋白表达水平,分析BTG1在不同组织中的表达差异及与胃癌病理特征及预后的关系。结果胃癌组织中的BTG1表达水平显著低于正常癌旁组织及健康正常胃黏膜组织,差异有统计学意义(P<0.01)。BTG1表达与性别、年龄、肿瘤大小及组织学分型等临床病理因素无关,而与胃癌TNM分期有关,具体表现在(Ⅲ+Ⅳ)期胃癌标本BTG1表达水平显著低于(Ⅰ+Ⅱ)期,差异有统计学意义(P<0.01)。进一步分析发现,BTG1低表达与上皮间质转化过程显著相关;且BTG1表达水平越低,预后5年生存率越差,差异有统计学意义(P<0.01)。结论 BTG1在胃癌中低表达,与转归及预后高度相关,可作为胃癌的生物学指标之一。

Objective To investigate the expressions and potential clinical significances of BTG1 on gastric cancer.Methods Forty-six tissue samples of gastric cancer and the corresponding adjacent normal tissue samples and thirty-one gastric tissues of healthy adults were collected.The expressions of BTG1 in different tissue samples were detected by qRT-PCR.The correlations between the BTG1 expressions and clinical features as well as prognosis and development of gastric cancer were analyzed.Results The levels of BTG1 expression in gastric cancer tissue were significant lower than that in the adjacent normal tissue samples and healthy samples（P0.01）.BTG1 was associated with TNM stages,but not with factors like age,sex,tumor size,tissue classification of gastric cancer;the levels of BTG1 in（Ⅲ ＋ Ⅳ） stages were higher than that in（Ⅰ ＋ Ⅱ） stages（P0.01）.The expressions of BTG1 were highly correlated with epithelium mesenchymal transition of gastric cancer;and were negatively correlated with the 5-year survival rate of human with gastric cancer（P0.01）.Conclusion The low expression of BTG1 in gastric cancer is highly correlated with prognosis and prognosis,which can be used as one of the biological indicators of gastric cancer.
