摘要
目的探讨细胞骨架结合蛋白4.1N在尤因肉瘤中的表达及其与细胞增殖、临床病理特征的关系。方法采用免疫组化En Vision法检测52例尤因肉瘤组织中4.1N的表达,分析4.1N表达与临床病理特征及预后的相关性,其中47例尤因肉瘤组织行Ki-67染色,并分析Ki-67与4.1N表达的相关性。结果 52例尤因肉瘤中4.1N的阳性率为35%(18/52),其中强阳性率仅占1.9%(1/52)。Ki-67增殖指数3%~85%。4.1N表达与Ki-67增殖指数呈负相关(P<0.05)。27例获得随访,Kaplan-Meier生存曲线显示,4.1N阴性患者的生存期明显低于4.1N阳性患者(P<0.05)。4.1N表达与患者年龄、性别、发病部位及转移无明显相关性。结论 4.1N在尤因肉瘤中主要呈表达缺失或低表达状态,其表达与Ki-67增殖指数呈负相关;4.1N可能参与尤因肉瘤细胞的增殖调控、并发挥抑癌基因的作用。此外,4.1N表达对尤因肉瘤患者预后具有一定预测作用。
Purpose To investigate the expression of cytoskeletal protein 4.1 N in Ewing's sarcoma and the correlation with cell proliferation as well as clinicopathological characteristics.Methods Immunohistochemical En Vision two-step staining was used to detect the expression of 4.1 N in 52 Ewing's sarcoma cases,which 47 cases of them were performed to detect the expression of Ki-67.The relationship between the expression of 4.1 N and Ki-67,and the correlation with the pathological features including prognosis were analyzed.Results The positive expression of 4.1 N was found in 35%(18/52) of Ewing's sarcoma.Lower frequency of 4.1 N strong expression(1.9%,1/52) was detected in the cohort.Ki-67 labeling index in Ewing's sarcoma varied from 3% to 85%.4.1 N expression displayed a negative correlation with Ki-67 index(P〈0.05).Among the follow-up of 27 cases,Kaplan-Meier survival analysis suggested that patients with 4.1 N-negative had shorter survival period than patients with 4.1 N positive expression(P〈0.05).However,the expression of 4.1 N was not statistically correlated with sex,age,tumor 's localization and metastasis.Conclusion 4.1 N mainly showed negative or lower expression in Ewing's sarcoma and was related to cell proliferation index Ki-67,which implied a potential function in regulating proliferation in Ewing's sarcoma.4.1 N expression has potentially predictive value in outcome of Ewing's sarcoma.
