摘要
目的探讨艾迪注射液对肺腺癌小鼠多西紫杉醇化疗敏感性的影响,并分析机制。方法取75只小鼠建立肺腺癌模型,分为5组:化疗组(2 mg/kg多西紫杉醇)、模型组(0.3 ml蒸馏水)、低剂量组(2 mg/kg多西紫杉醇+50 mg/kg艾迪注射液)、中剂量组(2 mg/kg多西紫杉醇+100 mg/kg艾迪注射液)、高剂量组(2 mg/kg多西紫杉醇+200mg/kg艾迪注射液),腹腔注射,每天1次。分别在1周后、2周后、3周后各处死5只,比较抑瘤率,组织凋亡抑制蛋白(Survivin)、B淋巴细胞瘤-2(Bcl-2)mRNA和蛋白的表达量。结果化疗组、低、中、高剂量组抑瘤率均随着时间的延长显著升高(P<0.05),且同一时刻各组比较,化疗组最低,低、高剂量组次之,中剂量组最高;化疗组、低、中、高剂量组Survivin、Bcl-2 mRNA和蛋白均随着时间的延长显著降低(P<0.05),且同一时刻各组比较,模型组最高,化疗组次之,低、高剂量组稍低,中剂量组最低。结论艾迪注射液对肺腺癌小鼠多西紫杉醇化疗敏感性有增强作用,能够改善肿瘤控制效果,以100 mg/kg剂量效果最佳,推测与下调Survivin、Bcl-2 mRNA和蛋白的表达有关。
Objective To explore the effect of Aidi injection on chemotherapy sensitivity of docetaxel in lung adenocarcinoma mice, and to analyze the mechanism. Methods Lung adenocarcinoma models were established in 75 mice, which were divided into 5 groups: chemotherapy group (2 mg/kg docetaxel), model group (0.3 ml distilled water), low dose group (2 mg/kg docetaxel+50 mg/kg Aidi injection), middle dose group (2 mg/kg docetaxel +100 mg/kg Addie injection), high dose group (2 mg/kg docetaxel+200 mg/kg Addie injection), by intraperitoneal injection, 1 time a day. After 1 week, 2 weeks, and 3 weeks, 5 mice were killed. The tumor inhibition rates, the expressions of apoptotic inhibitory protein (Survivin) and B lymphocyte tumor-2 (Bcl-2) mRNA and proteins were compared. Results The tumor inhibition rates in chemotherapy group, low, middle and high dose group increased significantly with time ( P 〈0.05), of which the chemotherapy group was the lowest at the same time, followed by the low and high dose group, the middle dose group highest. The expressions of Survivin, Bcl-2 mRNA and proteins in chemotherapy group, low, middle and high dose group decreased significantly with time ( P 〈0.05), of which the model group were the highest at the same time, followed by the chemotherapy group, low and high dose groups slightly lower, the middle dose group lowest. Conclusion Aidi injection has an enhanced effect on the chemosensitivity of docetaxel in lung adenocarcinoma mice, which can improve the effect of tumor control, and the effect of 100 mg/kg dose is best. It is presumed to be related to the down-regulation of Survivin, Bcl-2 mRNA and protein expressions.
