摘要
目的观察G_2/S期应答相关蛋白1(GTSE1)在胃癌和癌旁正常胃组织中的表达并探讨其意义。方法采用qRT-PCR和Western blotting法分别检测12例份新鲜胃癌和癌旁正常胃组织标本中GTSE1 mRNA和蛋白表达。免疫组化法检测56例胃癌患者癌组织及癌旁正常胃组织中GTSE1蛋白表达。分析GTSE1蛋白表达与胃癌患者临床病理特征的关系。COX回归模型分析胃癌患者预后的影响因素。结果与癌旁正常胃组织相比,GTSE1蛋白和mRNA在胃癌组织中表达升高(P均<0.05);GTSE1高表达与肿瘤大小、组织分化程度、肿瘤浸润深度、淋巴结转移、TNM分期有关(P均<0.05);COX回归模型分析显示,TNM分期和GTSE1表达是胃癌患者预后的独立影响因素(P均<0.05)。结论 GTSE1在胃癌组织中呈高表达,且GTSE1蛋白过表达与胃癌患者恶性病理特征及不良预后密切相关。
Objective To investigate the expression and clinical significance of G2 and S phase-expressed-1( GTSE1)in the gastric cancer( GC) tissues and its adjacent normal gastric tissues. Methods The expression of GTSE1 mRNA and protein in 12 cases of fresh GC tissues and adjacent normal gastric tissues were respectively detected by qRT-PCR and Western blotting. The expression of GTSE1 protein in 56 cases of GC tissues and adjacent normal gastric tissues was evaluated by immunohistochemistry. The relationship between the expression levels of GTSE1 protein and clinical pathological characteristics in patients with gastric cancer was analyzed. At last,the prognostic factors for GC patients was assessed by COX regression model. Results Compared with the adjacent normal gastric tissues,GTSE1 protein and mRNA were highly expressed in the gastric cancer tissues( both P〈0. 05). The high expression of GTSE1 protein was related to the tumor size,degree of differentiation,depth of tumor invasion,lymph node metastasis,and TNM staging( all P〈0. 05).COX regression model analysis manifested that TNM staging and GTSE1 expression were important independent prognostic factors for patients with gastric cancer( both P〈0. 05). Conclusion GTSE1 was highly expressed in the GC tissues,and the overexpression of GTSE1 protein is closely related to the malignant pathological features and poor prognosis.
