摘要
目的通过检测慢性阻塞性肺疾病(COPD)缺氧性脑损伤大鼠外周血中炎症因子和神经相关分子水平,探讨其变化和意义。方法通过大鼠被动吸烟建立COPD模型。采用酶联免疫法分析外周血中炎症因子[γ干扰素(IFN-γ)、IL-1β、IL-6、C反应蛋白(CRP)和TNF-α]水平,外周血神经相关分子[神经丝蛋白(NF)、神经特异性烯醇化酶(NSE)、Tau蛋白、脑钠素(BNP)、大鼠髓磷脂相关糖蛋白(MAG)、髓鞘碱性蛋白(MBP)和少突胶质细胞髓鞘糖蛋白(OMgp)]的水平。结果 COPD组大鼠外周血压炎症因子和外周血神经相关分子均升高。COPD组大鼠中MAG与IL-1β,BNP与IL-6和CRP均呈正相关(r=0.595、0.386、0.429,均P<0.05)。结论 COPD伴随慢性炎症反应,神经系统慢性炎症和缺氧可能导致神经细胞和胶质细胞受损。
Objective To investigate the peripheral blood levels of inflammatory factors and neural-related factors in rats with chronic obstructive pulmonary disease(COPD). Methods COPD model was induced by cigarette smoking in rats. The plasma levels of inflammatory factors IFN-γ, IL-1β, IL-6, C-reactive protein(CRP), and tumor necrosis factor-α(TNF-α) and neural-related molecules neurofilament,neuron-specific enolae(NSE), Tau protein, brain natriuretic peptide(BNP), myelin-associated glycoprotein(MAG), myelin basic protein, and oligodendrocyte myelin glycoprotein(OMG) were measured by enzyme linked immunosorbent assay(ELISA). Results Rat COPD model was successfully established. All the measured inflammatory factors and neurons and glia-released molecules in the plasma of COPD rats were significantly higher than those in normal rats. In COPD rats, MAG was positively correlated with IL-1β, while BNP was positively correlated with IL-6 and CRP. Conclusion The results indicate that inflammation exists in COPD, and the chronic inflammation and hypoxia may result in damages of neuron and glias in COPD rats.
