NK-lysin在人工感染绵羊肺腺瘤病毒肺组织的表达及其在不同品种绵羊的多态性分析

Expression of NK-lysin in lung tissues of sheep infected with pulmonary adenomatous virus and analysis of polymorphism in different sheep breeds

通过分析新疆不同品种绵羊NK-lysin基因多态性,探讨新疆不同品种绵羊NK-lysin基因多态性与抗病力的关系。构建了对建立的绵羊肺腺瘤病人工感染模型肺组织提取总RNA,经实时荧光定量PCR方法检测发病和健康绵羊肺组织中NK-lysin基因的表达量,对比不同病理条件下绵羊NK-lysin基因的表达差异。结果表明:萨福克羊和哈萨克羊NK-lysin基因存在多态性以及突变。通过荧光定量RT-PCR检测,感染绵羊肺腺瘤病肺组织NK-lysin的表达水平明显低于健康绵羊,说明Nk-Lysin参与绵羊肺腺瘤的病理形成,为绵羊抗肿瘤分子机制研究提供理论依据。

By analyzing the polymorphisnl of NK -lysin gene in different sheep breeds of Xinjiang, we discussed the relation- ship between NK- lysin gene polymorphisnl and disease resistance in Xinjiang sheep breeds. In addition, the total RNA was extracted from the lung tissue of the established sheep lung adenoma, and the expression of NK -lysin gene in the lung tissues of the healthy sheep was detected by real -time fluorescence quantitative PCR, and the difference in the expression of the NK - lysin gene in the sheep was compared under different pathological conditions. The results show that the existence of Suffolk sheep and Kazakstan sheep NK - lysin gene polymorphisnl and mutation. The expression of NK - lysin in lung adenoma ilffee- ted sheep was signifieamly lower lhan that of heallhy sheep by- fluorescence quanlitalive RT - PCR, indicaling lhal Nk - Lysin was involved in the pathological process of the sheep lung adenoma, and provided a theoretical basis for the study of the molec- ular mechanism of the sheepg anti - tumor molecular mechanism.
