金蛭方对缺血性中风大鼠caspase-3、Bax、Bcl-2蛋白表达的影响

Effects of Jinzhi fang on Expression of Caspase-3,Bax and Bcl-2 proteins in Rats with Ischemic Stroke

目的:通过观察金蛭方对缺血性中风大鼠缺血侧大脑皮质区细胞调亡相关因子Bax、Bcl-2、caspase-3蛋白含量的影响,探讨金蛭方治疗缺血性中风的作用机制。方法:采用线栓法复制缺血性中风大鼠模型,随机分为假手术组、模型对照组、金蛭方高剂量组、金蛭方低剂量组、阳性药尼莫地平片对照组。应用western-blot技术检测caspase-3、Bax、Bcl-2蛋白含量变化。结果:模型组大鼠缺血侧皮质区Bcl-2蛋白表达较假手术组明显降低(P<0.01);caspase-3、Bax蛋白表达较假手术组明显升高(P<0.01)。用药后各治疗组大鼠缺血侧皮质区Bcl-2蛋白表达明显升高(P<0.05,P<0.01);caspase-3、Bax蛋白表达明显降低(P<0.05,P<0.01)。高、低剂量组在增强Bcl-2蛋白表达和降低caspase-3、Bax蛋白表达方面的作用优于对照组。结论:金蛭方通过调节缺血性中风大鼠caspase-3、Bax、Bcl-2蛋白表达达到拮抗缺血性中风作用。

Objective To observe the effect of Jinzhifang on the content of Bax,bcl-2,caspase-3 protein in cerebral cortex region of ischemic stroke rats,and to investigate the mechanism of the treatment of ischemic stroke with Jinzhifang. Methods Using the suture method to copy the rat model of ischemic stroke were randomly divided into sham operation group,model control group,the Jinzhi fang decoction high dose group,the Jinzhi fang low dose group,positive drug nimodipine tablets in the Jinzhi fang. The content changes of caspase-3,Bax and bcl-2 were detected by using western-blot technique. Results The expression of Bcl-2 protein in ischemic cortex in model group was significantly lower than that in sham group（ P〈0. 01）. The expression of caspase-3 and Bax protein was significantly higher than that of sham operation group（ P 0. 01）. The expression of Bcl-2 protein in the ischemic cortex of rats increased obviously after the treatment（ P 0. 05,P 0. 01）. The expression of caspase-3 and Bax protein decreased obviously（ P 0. 05,P 0. 01）. The effect of Bcl-2 protein expression and decrease of caspase-3 and Bax protein in high and low dose group was better than that in control group.Conclusion The Jinzhi fang through regulation of ischemic stroke rat Expression of casase-3,Bax,Bcl-2 proteins and release in order to achieve the antagonistic effect of ischemic apoplexy.
