抑钙剂NPS 2143对创伤失血性休克大鼠心血管功能的影响 被引量:4

摘要 目的观察抑钙剂NPS 2143对创伤失血性休克(THS)大鼠心血管功能的影响。方法采用大鼠创伤失血性休克模型,观察NPS 2143输注对THS大鼠的存活情况、平均动脉血压与去甲肾上腺素诱导的升压反应、以及血流动力学指标的影响。结果未经治疗的大鼠在创伤失血性休克后数小时内全部死亡,单纯给予乳酸林格氏液(LR)复苏有轻度的改善作用,NPS 2143输注可明显延长THS大鼠的存活时间,提高24 h存活率。休克后大鼠的血压和去甲肾上腺素(NE)诱导的升压反应都显著降低,LR复苏有一定的恢复作用,NPS 2143能明显改善血压和升压反应。同时,NPS 2143可明显改善休克后降低的血流动力学指标,如左心室收缩压(LVSP)和左心室压力最大上升/下降速率(±dp/dt_(max)),其效果明显优于单纯LR复苏。结论抑钙剂NPS 2143对创伤失血性休克后心血管功能有明显的改善作用。 Objective The present study is to investigate the effects of calcilytics NPS 2143 on cardiovascular function after traumatic hemorrhagic shock( THS) in rats. Methods With a rat model of traumatic hemorrhagic shock,the effects of NPS 2143 on the animal survival,mean arterial blood pressure( MAP) and the pressor response of norepinephrine,and hemodynamic parameters were observed.Results All theuntreated THS rats died within several hours after shock. Fluid resuscitation with Lactated Ringer's( LR) solution slightly improved the survival outcomes. NPS 2143 infusion significantly prolonged the survival time and increased the 24-hour survival rate of THS rats. MAP and the pressor effect induced by norepinephrine( NE) were significantly reduced and resuscitation with LR showed some improvement after shock. NPS 2143 could significantly improve MAP and the pressor response of THS rats. Meanwhile,NPS 2143 could significantly improve the reduced hemodynamic parameters of THS rats,including left ventricular systolic pressure( LVSP) and the maximum rise/drop rate of left ventricular pressure( ± dp/dt_(max)). Its effect was significantly better than resuscitation purely with LR. Conclusion Calcilytics NPS 2143 showed a good effect of improvement in cardiovascular function after traumatic hemorrhagic shock.
