摘要 目的探讨SB203580抑制p38MAPK信号通路对足细胞特征性蛋白表达的影响。方法应用阿霉素体外刺激培养足细胞建立足细胞损伤模型,设置实验组和对照组,实验组加入p38MAPK信号通路抑制剂SB203580,对照组加入相等量的1640培养液,采用实时荧光定量PCR(Q-PCR)及Western blot检测足细胞特征性蛋白Nephrin和Podocalyxin的变化。结果与对照组的比较,实验组足细胞特征性蛋白Nephrin及Podocalyxin基因的mRNA和蛋白表达水平显著升高(P<0.05)。结论 SB203580能够将p38MAPK信号通路转导阻断,从而有效地抑制足细胞受到损伤,这对于临床干预儿童原发性肾病足细胞的病变是十分有利的,进而为避免蛋白尿的大量形成给予了十分关键的理论根据,具有重要的临床研究价值。 Objective To investigate the effect of a p38MAPK pathway inhibitor SB203580 on theinjury of podocyte.Methods Adriamycin was applied to stimulate podocyte in vitro as a control group.The experimental group were added SB203580 in order to inhibit p38MAPK signaling pathway on the basis of the control group.Real-time quantitative PCR(Q-PCR)and Western Blot(WB)were adopted to measure the expression of gene and protein of Nephrin and Podocalyxin.Results Compared with control group,the mRNA and protein of Nephrin and Podocalycin showed a higher expression(P〈0.05).Conclusion SB203580 could alleviate podocyte injury hrough inhibiting p38MAPK signaling pathway.The provide a vital theoretical basis on clinical intervention ofcytopathic effect.
