高效液相色谱法同时测定气管炎糖浆中6种有效成分

Simultaneous Determination of Six Ingredients in Qiguanyan Syrup by HPLC

目的建立同时测定气管炎糖浆中虎杖苷、白藜芦醇、柚皮苷、新橙皮苷、大黄素和大黄素甲醚含量的方法。方法采用高效液相色谱法,色谱柱为Inertsil ODS-SP C18,流动相为乙腈-0.1%磷酸溶液(梯度洗脱),流速为1.0 m L·min-1,检测波长分别为306 nm(虎杖苷、白藜芦醇)、280 nm(柚皮苷、新橙皮苷、大黄素和大黄素甲醚),柱温为30℃,进样量为5μL。结果虎杖苷、白藜芦醇、柚皮苷、新橙皮苷、大黄素、大黄素甲醚分别在36.32~726.4μg·m L^(-1)(r=0.999 4),2.338~46.75μg·m L^(-1)(r=0.999 9),4.147~82.94μg·m L^(-1)(r=0.999 9),5.310~106.2μg·m L^(-1)(r=0.999 9),1.602~32.03μg·m L^(-1)(r=0.999 9),0.102 5~2.050μg·m L^(-1)(r=0.999 9)范围内线性关系良好;精密度、稳定性、重复性实验的RSD<2.0%;平均回收率分别为97.2%(RSD=1.1%),96.0%(RSD=1.0%),97.8%(RSD=1.6%),96.2%(RSD=0.8%),101.6%(RSD=1.8%),100.8%(RSD=1.9%)。结论该方法灵敏、准确、可靠,可用于同时测定气管炎糖浆中6种成分。

Objective To establish a method for the simultaneous determination of polydatin,resveratrol,naringin,neohesperidin,emodin and physcion in Qiguanyan syrup. Methods HPLC method was used.The separation was performed on the column of Inertsil ODS-SP C18 with mobile phase of acetonitrile-0. 1% phosphoric acid solution（ gradient elution） at a flow rate of 1. 0 m L · min-1,the detection wavelength were set at 306 nm（ polydatin,resveratrol） and 280 nm（ naringin,neohesperidin,emodin and physcion）,column temperature was 30 ℃,and the injection volume was 5 μL. Results The linear range was 36.32-726.4 μg·m L^-1（ r = 0.999 4） for polydatin,2.338-46.75 μg·m L^-1（ r = 0.999 9） for resveratrol,4.147-82.94 μg·m L^-1（ r= 0.999 9） for naringin,5.310-106.2 μg·m L^-1（ r = 0.999 9） for neohesperidin,1.602-32.03 μg·m L^-1（ r= 0.999 9） for emodin,0.102 5-2.050 μg·m L^-1（ r = 0.999 9） for physcion.RSDs of precision,stability and reproducibility tests were less than 2.0%. The average recoveries were 97.2%（ RSD = 1.1%）,96.0%（ RSD = 1.0%）,97.8%（ RSD = 1.6%）,96.2%（ RSD = 0. 8%）,101. 6%（ RSD = 1. 8%） and 100. 8%（ RSD = 1. 9%） respectively. Conclusion The method was sensitive,accurate and reliable,and can be used for the simultaneous determination of six ingredients in Qiguanyan syrup.
