摘要
目的观察比较三伏灸不同灸量对哮喘型新西兰兔肺组织TGF-β1蛋白及mRNA表达的影响。进一步探讨TGF-β1在哮喘型新西兰兔气道重塑过程中发挥的作用。方法选体质量为(2.5±0.3)kg的雄性新西兰兔24只。用随机数字表随机分为空白组、模型组、实验组、对照组。空白组不造模,其余各组支气管哮喘气道重塑模型造模后,模型组不干预,实验组、对照组分别于"三伏天"以隔姜灸9壮与3壮灸量加中药贴敷法干预。实验结束后,取新西兰兔肺组织,检测肺组织TGF-β1蛋白表达及TGF-β1基因表达。结果与空白组比较,模型组、实验组、对照组的TGF-β1蛋白及mRNA表达明显升高(P<0.05);与模型组比较,实验组、对照组的TGF-β1蛋白及mRNA表达明显降低(P<0.05);与对照组比较,实验组TGF-β1蛋白及mRNA表达明显降低(P<0.05)。结论增加三伏隔姜灸量达到9壮可以更好地下调肺组织中TGF-β1蛋白及基因表达,通过抑制TGF-β1的合成,改善哮喘型新西兰兔气道重塑,从而有效缓解哮喘症状。
Objective To observe and compare the influence of ditterent amounts of dog day moxi- bustion on TGF -β1 protein and mRNA expression of the New Zealand rabbits with asthma. To thrther ex- plore the role of TGF-β1 in airway remodeling of New Zealand rabbits with asthmatic. Method 24 male New Zealand white rabbits were randomly divided into blank group, model group, control group, and ex- perimental group . Without the blank group, the airway remodeling model was established in the rest group. Model group only given airway remodeling without any treatment. Experimental group and the control group were given ginger moxibustion with 9 times and 3 times. At the end of the experiment, the expression of TGF-β1 protein and TGF-β1 gene expression in lung tissue were detected. Result Compared with the blank group, the protein and gene expression of TGF-β1 were significantly increased in the model group, control group and experimental group (P〈0.05). Compared with the model group, the protein and gene ex- pression of TGF-β1 were significantly decreased in the control group and experimental group (P〈0.05). Compared with the control group, the protein and gene expression of TGF-β1 were significantly decreased in experimental group (P 〈 0.05 ). Conclusion The increase of the amount of ginger moxibustion to 9 strong can improve the expression of TGF-β1 protein and gene in the lung tissue. By inhibiting the synthesis of TGF-β1, the airway remodeling of the asthma New Zealand rabbits can be improved and the symptoms of asthma can be ettectively relieved.
