艾灸对阳陵泉穴区皮肤组织五羟色胺和组胺的影响 被引量:7

Effect of Moxibustion on the Expressions of 5-Hydroxytryptamine and Histamine in Skin Tissue at Yanglingquan (GB34)

摘要 目的运用免疫组化结合共聚焦技术揭示艾灸对阳陵泉穴区皮肤组织五羟色胺(5-HT)、组胺(HA)的影响。方法以大鼠为实验对象,剃除双后肢阳陵泉穴区毛发,充分暴露皮肤组织,对左侧阳陵泉穴进行艾灸治疗,灸后即刻灌流,并分别取两侧穴区皮肤组织(1 cm×1.5 cm),经25%高渗糖脱水后用冰冻切片机制成20?m系列组织切片,然后分别用5-HT及HA抗体进行荧光免疫组化检测。结果艾灸后皮肤组织中5-HT和HA阳性表达明显增强,5-HT分别在表皮层与真皮层之间,皮下组织层沿血管形成连续的增强带;HA主要集中分布在真皮层深部及皮下组织的血管旁。通过显微镜计数,艾灸后皮肤组织中5-HT和HA阳性标记数量明显高于正常皮肤组织,差异均具有统计学意义(P<0.05)。结论艾灸能够使阳陵泉穴区皮肤组织中5-HT和HA的表达上调,其可能参与艾灸作用通路的信息传递。 Objective To investigate the influence of moxibustion on the expressions of 5-hydroxytryptamine(5-HT) and histamine(HA) in skin tissue at Yanglingquan(GB34) by using immunohistochemistry combined with confocal technique. Method The rat was used in this experiment. The skin tissue on hind limbs Yanglingquan(GB34) was exposed by shaving. Subsequently, moxibustion was carried out on the left side of Yanglingquan(GB34). After moxibustion, the local skin tissue of 1 cm × 1.5 cm on bilateral Yanglingquan(GB34) of rats were collected, and put into 25% sucrose solution. The tissues were sliced at 20 ?m on a cryostat. The 5-HT and HA antibodies were used for fluorescent immunohistochemical test. Result The positive expressions of 5-HT and HA increased in the local skin tissue after moxibustion, in which 5-HT increased between the epidermis and dermis, in subcutaneous tissue layer forming a continuous band of enhancement along the blood vessels, while HA was majorly distributed nearby the blood vessels of deep dermis and subcutaneous tissue. After counting the positively-expressed 5-HT and HA by microscopic field of view, the numbers of 5-HT and HA positive markers were significantly lager in post-moxibustion skin tissue than in normal skin tissue. The differences were statistically significant(P〈0.05). Conclusion Moxibustion plays a role in up-regulating the level of 5-HT and HA in the local skin tissue at Yanglingquan(GB34). It suggests that 5-HT and HA may be involved in the signal transduction of moxibustion-related pathways.
