垂体瘤病灶内LRIG1、Gadd45g表达量与肿瘤细胞自噬、凋亡、侵袭的相关性研究 被引量:2

摘要 目的:研究垂体瘤病灶内LRIG1、Gadd45g表达量与肿瘤细胞自噬、凋亡、侵袭的相关性。方法:选择2014年6月~2017年12月期间在我院接受手术切除的垂体瘤患者并留取侵袭性垂体瘤组织、非侵袭性垂体瘤组织,另取同期因颅脑外伤在我院接受手术治疗的患者作为对照并留取正常脑组织;测定组织样本中LRIG1、Gadd45g及自噬、凋亡、侵袭基因的mRNA表达量。结果:侵袭性垂体瘤病灶和非侵袭性垂体瘤病灶组织中LRIG1、Gadd45g、Beclin1、LC3-Ⅱ、Bax、NKG2D、E-cadherin的mRNA表达量均显著低于正常脑组织,PTTG1、c-Myc、Gal-3、Bcl-2、EphA2、HSP27、MMP14、VEGF的mRNA表达量均显著高于正常脑组织且侵袭性垂体瘤病灶组织中LRIG1、Gadd45g、Beclin1、LC3-Ⅱ、Bax、NKG2DE-cadherin的mRNA表达量均显著低于非侵袭性垂体瘤病灶组织,PTTG1、c-Myc、Gal-3、Bcl-2、EphA2、HSP27、MMP14、VEGF的mRNA表达量均显著高于非侵袭性垂体瘤病灶组织;LRIG1、Gadd45g的mRNA表达量与Beclin1、LC3-Ⅱ、Bax、NKG2D、E-cadherin的mRNA表达量呈正相关,与PTTG1、c-Myc、Gal-3、Bcl-2、EphA2、HSP27、MMP14、VEGF的mRNA表达量呈负相关。结论:垂体瘤病灶内LRIG1、Gadd45g的低表达能够抑制肿瘤细胞的自噬及凋亡、促进肿瘤细胞的侵袭。 Objective:To study the correlation of LRIG1 and Gadd45 g expression in pituitary tumor with the autophagy,apoptosis and invasion of tumor cells.Methods:Patients with pituitary tumor who underwent surgical resection in our hospital between June 2014 and December 2017 were selected,and the invasive pituitary tumor tissues and noninvasive pituitary tumor tissues were collected;patients who underwent surgery for craniocerebral trauma in our hospital during the same period were selected as the control group,and normal brain tissues were collected;the mRNA expression levels of LRIG1,Gadd45 g as well as autophagy,apoptosis and invasion genes in tissue samples were measured.Results:LRIG1,Gadd45 g,Beclin1,LC3-Ⅱ,Bax,NKG2 Dand E-cadherin mRNA expression in invasive pituitary tumor and noninvasive pituitary tumor tissues were significantly(P〈0.05)lower than those in normal brain tissues whereas PTTG1,c-Myc,Gal-3,Bcl-2,EphA2,HSP27,MMP14 and VEGF mRNA expression were significantly(P〈0.05)higher than those in normal brain tissues,and LRIG1,Gadd45 g,Beclin1,LC3-Ⅱ,Bax,NKG2 Dand E-cadherin mRNA expression in invasive pituitary tumor tissues were significantly(P〈0.05)lower than those in noninvasive pituitary tumor tissues whereas PTTG1,c-Myc,Gal-3,Bcl-2,EphA2,HSP27,MMP14 and VEGF mRNA expression were significantly(P〈0.05)higher than those in noninvasive pituitary tumor tissues;LRIG1 and Gadd45 g mRNA expression were positively correlated with Beclin1,LC3-Ⅱ,Bax,NKG2 Dand E-cadherin mRNA expression,and negatively correlated with PTTG1,c-Myc,Gal-3,Bcl-2,EphA2,HSP27,MMP14 and VEGF mRNA expression.Conclusion:The low expression of LRIG1 and Gadd45 g in pituitary tumor can inhibit the autophagy and apoptosis of tumor cells and promote the invasion of tumor cells.
