猪源肺炎克雷伯菌的分离鉴定及生物学分析

Isolation,identification and biological characteristics analysis of Klebsidla pneumoniae from swine

为了对河南省某猪场疑似细菌感染病例进行确诊,采集了病猪肠管中的样品并从中分离到1株革兰阴性短杆菌,对其进行了培养特性观察、生化试验、药敏试验以及16S r RNA基因序列分析.结果显示:分离菌的生化特性、培养特性基本符合肺炎克雷伯菌特征,16S r RNA基因序列与肺炎克雷伯菌的同源性均在98%以上,因此,将该分离菌株命名为Kpn;序列分析表明该分离菌株与猪源肺炎克雷伯菌NR114507的同源性最高,与大肠杆菌、普通变形杆菌、胸膜肺炎放线杆菌的同源性相对较低;分离株具有较强的致病性和多重耐药性,对美洛西林、哌拉西林、强力霉素、美满霉素、头孢哌酮敏感,但对氨苄西林、先锋霉素IV、先锋霉素V、先锋霉素VI、卡那霉素、新霉素等多种药物耐药.研究为猪肺炎克雷伯菌的分离鉴定及治疗提供了参考.

In order to diagnosis the suspected bacterial infection in a pig farm in Henan Province,the sample of the sick pig＇s intestinal was collected and 1 strains of gram negative bacillus was isolated.The culturel characteristics,biochemical properties test,drug sensitivity test and 16 S r RNA gene sequence analysis were carried out.The results showed that：The biochemical characteristics of the isolated bacteria were in accordance with Klebsidla pneumonia,the homology of 16 S r RNA gene sequence with Klebsidla pneumoniae was above 98%.The isolated strain was designated as Kpn.Sequence analysis showed that the isolated strain had the highest homology with Klebsidla pneumoniae NR114507,its homology with Escherichia coli,Proteusb vulgaris and actinobacillus pleuropneumoniae was relatively low.The isolated strain was pathogenic and multi-drug resistance was strong,especially to ampicillin,cephalosporin（IV,V and VI）,kanamycin,neomycin.But it was sensitive to Melocillin,piperacillin,doxycycline,fulminomycin, cefoperazone.This study provides a reference for the isolation,identification and treatment of klebsiella pneumoniae from pigs.
